| Cordycepin,one of the important compounds in Cordyceps militaris,has the activities of anticancer,antimicrobial,anti-inflammatory,and soon.Cordyceps sinensis as a highly medicinal fungi,in the face of growing demand and increasingly decimated wild Cordyceps resources,artificial domestication of wild Cordyceps,artificial cultivation and submerged fermentation is an effective means to replace the wild Cordyceps lacking.In this study,the use of grain produce cordycepin,ensure food security;by optimization of medium,optimization of culture conditions and screening of growth factors,and establish kinetic model.According to Cordyceps militaris nutritional needs,study the impact of plant quality carbon,nitrogen,type and concentration of cordycepin production,determine the appropriate Cordyceps militaris FFCC5111 fermentation medium cordycepin: 12° wort of protein hydrolysis time 80 min.Research the lighting conditions and ventilation on cordycepin synthesis.Found that prolonged light time in favor of cell growth;and cordycepin production show the first increased and then decreased,the maximum light time for 16h/d,cordycepin production was 109.3 mg/L.Meanwhile,the increase in ventilation can significantly promote the mycelial growth and cordycepin of synthesis.When the day pass into the equivalent of 25 times the volume of the culture medium of sterile air,biomass and the yield of cordycepin is highest,which cordycepin yield 50.8 mg/L,is 3.15 times the static culture.Investigation wort sugar content,the amount of sugar supplement and time of sugar supplement affect for C.militaris FFCC5111 cordycepin production.Variance analysis show that the sugar content of wort achieve highly significant;amount of supplement sugar and time of supplement sugar were not significantly impact.The effects of different additives on cordycepin production by submerged culture of Cordyceps militaris were investigated.Results indicated that the addition of adenosine,Phenylalanine,adenine,glycine or glutamine could result in a large improvement in cordycepin production by submerged fermentation of C.militaris,especially the addition of 1.5g/L adenosine,which resulted in a cordycepin production of 772.5 mg/L,exhibiting a 9.39-fold increase than that obtained in the basal medium culture especially.Adopting 12 ° beerwort which add 1.5 g/L adenosine as a medium in 50 L batch system,the kinetic model for C.militaris FFCC 5111 was based on the Logistic and Luedeking-Piret equations for cell growth,product formation.MATLAB was utilized to solve the model parameters and verify the model fitting result.These results generally reflected the kinetics feature of the batch fermentation of C.militaris. |
