Study On The Synthesis And Antitumor Activity Of Ganoderma Lucidum Selenium Polysaccharide

Ganoderma Lucidum Karst,which refers to the polyporaceae family plant,is the whole plant of Ganoderma sinense or Ganoderma lucidum.As a rare traditional Chinese medicine,Ganoderma Lucidum Karst is known as auspicious grass and immortal grass,and there are thousands years for it's medicinal history.In which,Ganoderma lucidum polysaccharides is one of it's many effective components,and has immunoregulation,antitumor,antioxidation,anti-aging,hypolycemic,reducing blood lipid effects.In recent years,pharmacological studies suggest that the pharmacological activity of selenium polysaccharide is more significant than polysaccharide and selenium,and it could be absorbed rapidly,without side effects.And it has a good effect on anti-heavy metal,antioxidation,antivirus,antitumor,immunoregulation and reducing the toxicity of other drugs chemotherapy.At present,lots of researches on Ganoderma lucidum polysaccharides was reported in China and abroad.But it is rarely reported for it that esterifying modify it to introduce the C-Se structure.The Ganoderma Lucidum used in this study was come from Gongcheng County,Guilin City,Guangxi Province.Firstly the author used the modern separation and analytical techniques to extract,separate and purification Ganoderma lucidum polysaccharides;then the purified part was esterifying modified to introduce the C-Se structure,and it was identified and characterized that the structures of acetylation/non-acetylation modification polysaccharides;last the author studied the antitumor activity in vitro of them.This paper was composed of two parts.The first part was an overview of Ganoderma lucidum polysaccharides and selenium polysaccharides.The second part was the experimental section,which included extraction,separation,purification,preliminary structure analysis of polysaccharide and synthesis,antitumor activity of selenium polysaccharide.The main contents of experimental section included the following five aspects:(1)The crude polysaccharides were obtained with the water extraction and alcohol precipitation method,and the protein was removed with the enzymatic binding Savage method.The effect of deproteinization was identified by the combined use of UV spectrophotometer and coomassie brilliant blue G-250.The protein content of removal protein polysaccharides is only 1.60%.(2)To obtain homogeneity components polysaccharides(GLP),the deproteinization polysaccharides were decolored with hydrogen peroxide method,removed the small molecule by dialysis,and purified by stepwise eluting with dextran gel.Its homogeneity was proved by specific optical rotation and gel chromatography,and its relative molecular weight was determined to be 4.344×104.(3)The monosaccharide composition of was measured by High Performance Liquid Chromatography(HPLC)after 3-Methyl-1-phenyl-2-pyrazolin-5-one(PMP)derivatization.It was characterized by IR and 13C-NMR.The results showed that GLP,which consists of mannose,glucose,galactose and fucose in the proportion of 2.40:12.46:7.17:1,is a homogeneous ?-pyran polysaccharide.(4)After esterifying modified GLP to introduce the C-Se structure and characterized by IR,the author synthesized two kinds of selenium polysaccharides: propionate selenium polysaccharide(PSGLP)and acetic acid selenium polysaccharide(ASGLP).It is 7.6 mg/g and 20 mg/g that the selenium content of their.(5)The antitumor activity in vitro of PSGLP and ASGLP was tested by MTT staining with Oxaliplatin,selenium and GLP as reference substance.The results showed that the two selenium polysaccharides had good anti-cancer activity for Hela(human cervical carcinoma cells)?CNE-2(Human nasopharyngeal carcinoma cells)?7721(human hepatoma cells).
