Apatite is a kind of calcium salt content material widely distributed in nature, with stable chemical properties, good surface chemical properties and outstanding performance of ion exchange. It has access to be used in hard tissue repair, medical release, inorganic antibacterial agent, biological separation, water treatment, catalytic, etc. According to the types of the structure of anion, apatite can be divided into different subspecies of mineral such as fluorinated, chlorinated, hydroxyapatite, etc. With similar chemical composition of hard tissue and crystal structure to human body leading to excellent bioactivity and biocompatibility, hydroxyapatite is the hot topic in the study of biological material.In this paper, we prepare a new kind of hydroxyapatite consisting of rod-like aggregates, and test its application in biological separation medium. In addition, we try to synthesis hydroxyapatite with different shells as raw material including fan shell and clam shell. Details are as follows:1. Using commercially available calcium salt and phosphate as raw materials, we explore a new way to prepare hydroxyapatite by homogeneous precipitation method. Firstly calcium-ion and phosphate-ion homogeneous solution was prepared, then mixed to be the precursor of final product.Thirdly pH of the system was controlled by dissolved urea due to hot water bath, and the precursor was changed into final product. Compared with the traditional preparation method, it is easy to control the whole process and the morphology of product is novel.2. Aiming to recycling and reusing waste, we use some common leftovers on the table as the source of calcium in the preparation of hydroxyapatite, including scallop shell and clam shell. The shells was changed to powder by wet ball mill, washing, drying and other processing, and then dissolved as calcium-ion solution by concentrated hydrochloric acid. Finally the calcium-ion solution was mixed with phosphate-ion solution which changed to hydroxyapatite along with urea dissolved in water bath. The product is of stable property, low cost, and the commercial value is considerable.3. The development of genetic engineering recently made gene expression more simple, but the separation of biomolecules developed relatively slowly. This section aims to use the prepared hydroxyapatite powder as column chromatography medium used in protein extraction, and evaluate the performance from the pressure of the column, the separation effect of proteins so as to explore its application in biological separation. The results showed that the synthesized hydroxyapatite was good at absorbing biological molecules such as DNA and protein, owned excellent pressure resistance and biological separation effect.
