Vitamin D3?vitamin D3,VD3?plays an important role in animals and human reproduction.VD3 can produce a series of metabolites catalyzed by vitamin D synthetases?CYP27A1 and CYP27B1?and degradation enzyme.1 alpha,25?OH?2D3?1,25 D3?which is the active form of VD3 and play its biological role mainly through combination of vitamin D receptor?VDR?.Studies show that adult mice,rats and people reproductive system have VDR and vitamin D synthetases expression.However,there is no study on the changes of VDR and vitamin D synthetases expression in the reproductive organs during the development of animal reproductive system and in the breeding and without breeding animals.In this study,male and female SD rats with different growth and development stages and5-month breeding and without breeding were studied to analyze the organ index of their main organs,detect the histomorphologic changes of reproductive organs by H&E staining method,check VDR and vitamin D synthetases CYP27A1,CYP27B1 mRNA expression changes by real-time quantitative PCR?real-time qPCR?technology,investigate the pattern of VDR and vitamin D synthase mRNA expression changes in the reproductive system organs of SD rats with different growth and development periods and of 5M breeding and without breeding SD rats.Further the mesenchymal stem cells?MSC?of 12M and 24M testis were isolated and cultured,and the expression changes of VDR,CYP27A1 and CYP27B1 mRNA were detected.The results showed that with the increase of age,both male and female rats reproductive organ index gradually increased,and began to decline after aging.The organs of male and female reproductive system in rats became mature with the development process,and the morphology began to be abnormal after aging.The reproductive system organs of SD rats with different growth and development periods and 5M breeding and without breeding SD rats has the expression of VDR and vitamin D synthetases CYP27A1 and CYP27B1 mRNA,and the expression presented a certain pattern.CYP27A1 mRNA expression is very significantly higher than CYP27B1 mRNA in the same period of reproductive system organs of SD rats with different growth and development periods and 5M breeding and without breeding SD rats.VDR,CYP27B1 and CYP27A1 mRNA of 24M testicular MSC was significantly higher than 12M testicular MSC.The research results demonstrated that VD3 may be synthesized into active VD3 by vitamin D synthetases and combined with the VDR,so as to exert its biological function in the reproductive system organs of SD rats with different growth and development periods and testicular MSC.Active VD3 metabolism and its biological effect presented a certain pattern in the process of SD rats reproductive organs growth and development.CYP27A1 catalytic product 25D3 synthesis into active VD3 by CYP27B1 perhaps is the main limiting step of active VD3 synthesis in the reproductive system organs of SD rats with different growth and development periods and testicular MSC,which limit the production of VD3 and its biological function.Breeding may has certain effects on the production of VD3 and its biological role in the reproductive system of female and male SD rats.The production of active VD3 and its biological effect were stronger in 24M testicular MSC than 12m testicular MSC.
