ObjectiveThe aims of this study were to determine the effects of?-1,3-galactosyltransferase 2(B3galt2)gene on learning and memory,neurons and synapses in mouse hippocampus,and to provide theoretical basis for further investigating the important role of B3galt2 and other glycosyltransferases in the central nervous system(CNS).MethodsB3galt2 mice were bred and genotyped using PCR.The Morris water maze was used to explore the learning and memory function.The expression pattern of B3galt2 gene in hippocampus was determined by x-gal(5-Bromo-4-Chloro-3-Indoiy ?-D-Galactopyranoside)staining.Immunohistochemistry study was used to observe the morphology of hippocampal neurons and synapses.Results1.6-month-old wild-type,B3galt2 heterozygous and B3galt2 homozygous mice from the same litter and the same gender were successfully obtained by breeding and genotyping.2.?-1,3-galactosyltransferase 2 gene knockout could induce theimpairement of learning and memory.3.The expression of ?-1,3-galactosyltransferase 2 gene in the hippocampus of the 6-month-old mouse brain was confirmed.4.The numbers of neurons in DG and CA1 of hippocampus,the expression of pre-synaptic marker SNAP25 and dendritic marker MAP2 were significantly decreased in B3galt2 heterozygous and homozygous mice.Conclusions1.?-1,3-galactosyltransferase 2 gene was expressed in the hippocampus of 6-month-old mouse brain.2.?-1,3-galactosyltransferase 2 might play important roles in learning and memory,neuronal function and synaptic plasticity in mouse hippocampus.
