Circular RNA(CircRNA)is a new class of novel and special non coding RNA molecules that was found in recent years.It is usually produced by a special alternative splicing mechanism that contains more than one exon to form circular RNA.CircRNA is almost present in all species and most of they are present in the cytoplasm of eukaryotic cells.In the present study,it is indicated that the expression of circRNA in human or animals has tissue specificity and disease specificity,and it is likely to be a new molecular marker.In the past,RNA was considered to be a linear molecule,but the found of circRNA was to increase people's new understanding of the diversity of RNA structure.Because the circRNA is not degraded by the RNA enzymes and it has stronger stability than the linear RNA,so it is more easier and more reliable to separate,enrichment,identification and detect circRNA.CircRNA and other types of small RNA(microRNA,miRNA)can make interaction with each other,it can also occur in the interactions between CircRNA-miRNA-mRNA to form a variety of network effects,exerts its biological functions and exert its biological function.The reason why these interactions occur is that the circRNA molecule contains miRNA binding sites.Once they interact on each other,it can counteract the inhibitory effect of miRNA on the target gene and increase the expression level of the target gene.Because some miRNAs are closely related to the disease,circRNA has a regulatory role in the occurrence and development of the disease.With the development of high throughput sequencing technology and bioinformatics,the circRNA has been identified in a variety of eukaryotic organisms.The basic research and application research of circRNA become a new hot spot now.But so far,there are few studies on the circRNA in plants,and there is also still no large scale study in plants.Based on the research of circRNA in the past,his project is make full use of RNA-seq datas and bioinformatics tools to make a deep and detailed study on the circRNA in Arabidopsis thaliana.We have achieved the following important results:1)Using Arabidopsis thaliana transcriptome data(experimental group and control group)after processing by RNase-R,we use the prediction method of Zhengyun_find_circ that generated by zhengyun,we totally found of 2310 ecircRNAs and the subsequent analysis was carried out.2)The basic features of RNA in Arabidopsis thaliana were analyzed,The circRNA formed by the 4 exons is the most.The second is the condition of 3 exons and 5 exons.and the number of circRNA was significantly decreased with the increase of the number of exons.in this study,we predict that one circRNA has a maximum of 75 exons.The number of circRNA formed from the second exon was the most,and the number of circRNA was less with the increase of the position of the exon.The formation of circRNA needs to reach a certain length.It can be inferred that the sequence of exon is too short to form circRNA.In addition,the statistical distribution of the length of the circRNA in Arabidopsis was done,the shortest one is 102bp,the median is 1144bp,,the longest is 11709bp,it is so that the circRNA can be regarded as a chain of long non coding RNA.The number of transcripts that form 2310 circRNAs is only 1110,which indicates that a gene may forme a plurality of circRNA.3)Combining the differential expression analysis results of RNA-seq data sets from 8 samples(col_RNase,m55_RNase),we discovered that the 8 samples were screened out of 622 differentially expressed circRNAs,Accounted for 26.93%(622/2310)of the entire candidate circRNA.under these 8 samples,with 120 and 502 show significant upregulation and downregulation.M55 mutations may affect the expression of circRNA,but its specific regulatory mechanisms and possible biological functions have yet to be verified.4)GO annotation analysis of transcripts of the circRNA was done,and it is found that most of the genes in the cellular components are mainly cells(cell,part cell)and cell organelles(organelle)components;Molecular functions include catalytic activity,binding proteins,etc.circRNA play an important role in the process and metabolism of cells in biological processes.It is indicated that the source gene of circRNA is widely distributed and has a wide variety of biological functions.It has important significance in the regulation of gene transcription.
