Tartary buckwheat(Fagopyrum tataricum)is one of an important cultivated crop in genus Fagopyrum,Polygonaceae.It is an important edible and medicinal plant,which has significant curative effect on cardiovascular disease,glucose decreases,immunity enhancement,anticancer agents,oxidation resistance,liver protection,and antianaphylaxis,etc.This experiment uses tetraploid tartary buckwheat seeds(1307-1178)as experimental material for extracting.First,three common macroporous resins were chosen to conduct separation and purification on the extracting solution of tetraploid tartary buckwheat to detect the difference of dynamic adsorption function among resins,and among them,the best resin suitable for the general flavone ingredient purification on tartary buckwheat can be confirmed.On this basis,the concentration of extract,pH value,flow rate,amount of water for washing,ethanol concentration,amount of ethanol were studied by dynamic adsorption,in order to determine the optimum purity technics parameters.The results showed that D-101 macroporous resin has the best adsorptive property.Its best adsorption condition is: the pH value of 4.0,the adsorption flow rate of 4BV/h and the concentration of buckwheat extracting solution of 0.3g/mL.And its best eluting condition is: water amount of 80 mL,ethanol concentration of 50%,and ethanol amount of 80 mL.Through this process,the purity of total flavonoids in standard tartary buckwheat extracts is about 3 times than before purification,indicating a good process for the separation and purification of the general flavone in tartary buckwheat seeds.A HPLC detection method was set up for testing the rutin content in tetraploid tartary buckwheat standardized extract.The chromatographic conditions,extraction process and methodology were investigated.The results showed that methanol is the best solvent,and that the ultrasonic extraction for 20 min is quite proper.Its chromatographic condition is: the wavelength is 360 nm,methyl alcohol: water = 65:35 as mobile phase system,Dumas C18 as experiment column,30? column temperature as the examine temperature.This method is accurate,stable and reliable for the rutin content in standardized extracts on tetraploid tartary buckwheat.Four models of antioxidant in vitro on ABTS· free radical,DPPH· free radical,·OH free radical and O2-·free radical were set for investigating the antioxidant capacity of tetraploid tartary buckwheat extractive,as comparison of Vc as the reference substance.The results showed that all tetraploid tartary buckwheat extract have antioxidant activity,and the activity scavenging free radical is as the order: Vc > standard extract > crude extract,the eliminating activity of the standard extract is about 2 times than the crude extract,indicating a better tetraploid tartary buckwheat standard extract than the crude extract,which can be used in Chinese herbal medicine as raw materials and in broadening the product form of traditional Chinese medicine.
