Plant molecular systematics has been widely used in various taxa on the research of the botany,reflects the certain application prospect and theoretical significance.Paeonia sect.Moutan DC is the main resources in China,the author of "the king of flowers" the laudatory name,has rich germplasm resources.With the development of tree peony breeding and the extensive cultivation of globalization,especially complicated relationships between species,only based on the traditional morphology is very difficult to accurate judgment to the related species and varieties,therefore needs to be reliable evidence of molecular biology to elucidate the peony group of system evolution and genetic relationship.This study with 9 wild species,60 cultivars of Paeonia rockii,and 13 accessions of Paeonia suffriticosa,3 foreign varieties as materials,through a leaf total DNA extraction,PCR amplification reaction.For genetic relationship between wild and cultivated varieties and the relationship between system evolution,orientation for the future breeding and the breeding of new varieties,and provide basic data for the development and utilization of germplasm,The results of the study are as follows:1.Based on the multiple karyogene and screening of chloroplast genome sequences,Finally got ITS karyogene and chloroplast gene mat K could identify the optimal gene sequence as sect.Moutan DC,Its both the optimal annealing temperature of 60 ? and 42.8 ? respectively.Using molecular markers about ITS and mat K could identify as sect.Moutan DC of interspecific relationship,the 9 wild species can be divided into two subgroups by both markers,Subsect Vagiatae and Subsect Succulent.Subsect Vagiatae including: Paeonia jishanensis,Paeonia rockii,Paeonia qiui,Paeonia ostii,Paeonia decomposita.The Paeonia rockii with Paeonia jishanensis and Paeonia ostii is closest relative,and with Paeonia qiui are closed genetic relationship,Paeonia decomposita is at the bottom of Subsect Vagiatae.Subsect Succulent including: Paeonia ludlowii,and the three species Paeonia lutea,Paeonia potaninii,Paeonia delavayi.3.About the relation of wild species and cultivated varieties:By the results of genetic markers on ITS and mat K are shown: wild Paeonia rockii has closely relationship with P.rockii “Fencuiqiu”,P.rockii “Fenxiuqiu”,P.rockii “Yuloucangjiao” and Paeonia ostii is close to cultivars of P.rockii “Zhuangyuanhong”,P.rockii “Zihelian”,P.rockii “Hongxiuqiu”.Paeonia rockii varieties for multi-line hybrid origin,and not by direct domestication of wild species,may be on the basis of the direct introduction of some varieties,through repeated hybridization and selection,The origin of the Paeonia rockii in xibei Paeonia suffruticosa,Paeonia ostii is also involved.4.About the relationship between cultivars:On the basis of ITS maker,the maker of mat K of clustering results could identify can be more intuitive that the cultivars relationship,From the zhongyuan Paeonia suffruticosa and the xibei Paeonia suffruticosa is roughly relatively concentrated together,show the same origin of Paeonia suffruticosa has closed genetic relationship.And within zhongyuan Paeonia suffruticosa and the xibei Paeonia suffruticosa,The same of color is similar to get together,showed that genetic relationship and color has certain relevance.Zhongyuan Paeonia suffruticosa and Paeonia rockii cultivars has crossed clustering phenomenon,through the analysis to get: Zhongyuan Paeonia suffruticosa,to a certain extent,involved in the formation of the varieties of Paeonia rockii.
