With the increasing development of microbial genome sequencing,a large number of microbial genomes have been accumulated. It is a very important task for bioinformatics to analyze gene structures, mine the biological meaning in the DNA sequences, extract the feature parameters and propose the algorithm of gene identification, moreover, the algorithm can be applied to annotate the genomes. In the thesis,I am devoted to set up a new algorithm for predicting the translation initiation sites of prokaryotic genes. The main work includes: studying the characteristics of the flanking sequences around translation initiation sites of prokaryotic genes, extracting feature parameters from the flanking sequences, and proposing the algorithm for identifying translation initiation sites based on the feature parameters. The algorithm has been employed to identify genes in prokaryotic genomes. The thesis can be divided into five parts:The first chapter introduces the background of the development of bioinformatics and main research results and innovations of the thesis. The second chapter presents the biology and mathematics knowledge, and some software used in this paper, which are related to the research task in the thesis. The third chapter is the main work in the thesis, which includes the parameter, the algorithm for identifying translation initiation sites of prokaryotic genes. The innovation points is that the predicted genes are firstly divided into the signal peptide genes and the non-signal peptide gene based on the N-terminal peptides of proteins, the feature parameters in two sets of prokaryotic genes are extracted to identify translation initiation sites, respectively. It is shown that the prediction results are highly accurate, especially in short genes. The fourth chapter is system analysis and in-depth discussions to the results of identifying translation initiation sites in prokaryotic genes. The fifth chapter is the conclusion part.First of all,the work in the thesis is simply summarized. The open questions are pointed out in the gene identification field. Finally, puts forward three main innovation points.
