As an efficient and versatile investigation approach in the field of biological and medical science,fluorescence cell imaging not only attracts academic attention but also contributes to potential applications.They can adapt to di erent experimental conditions and possess tunable optical properties.Nowadays,many kinds of organic dyes,fluorescent proteins,and quantum dots(QDs)are commercially available for cell imaging.However,some limitations of these current fluorescent materials have become obstacles restricting their applications.For example,some organic dyes suffer from photobleaching and cellular toxicity,fluorescent proteins are often associated with laborious and lengthy procedures,and although QDs have superior brightness and photostability,their cytotoxicity remains a challenge for further applications in vivo.With unique photophysical properties and good biocompatibility,conjugated polyelectrolyte(CPEs)gradually attracted biologists and materials scientists' attention and widely used in the field of biological and chemical sensors.This paper designed and synthesized three novel conjugated polyelectrolyte,which successfully applied to cell imaging and the detection of ATP.The main contents are as follows:1.Different to CPEs with the poly(phenylene ethynylene)backbone,we introduced terthienyl to the backbone of conjugated polymer,and modified an positively charged groups in the side chain to give three cationic conjugated polyelectrolytes,which a have a common backbone and side chains of di erent lengths.Bseides,their synthetic methods and photophysical properties were described.2.Selecting ATP,ADP,AMP and eight other anions as an analyte,we studied its interaction with PPET3-N1,PPET3-N2 and PPET3-N3.We found the spectroscopic properties of three conjugated polyelectrolyte are a ected by ATP,characterized by substantially fluorescence intensity decreases.Compared to PPET3-N1 and PPET3-N3,PPET3-N2 get a greatest response to ATP.3.The application of three polymers in cell imaging was researched by confocal fluorescence microscopy.The result demonstrated all of three conjugated polyelectrolyte showing bright fluorescence were mainly localized on the cell membrane and inside of cells in a measure.Besides,PPET3-N2 showed the most great response to ATP.We use it as a probe to successfully detect the ATP which localized on the cell membrane by means of cell imaging.The result indicated that PPET3-N2 has potential applications in cell imaging and ATP semiquantification in vivo.In addition,the interaction of three conjugated polyelectrolyte with phospholipid or surfactant was also studied.
