Floral induction is an importment process in high plant,and CONSTANS gene is a key regulator in this process.In the previous studies,16 classes CONSTANS homologous genes from mango(Mangifera indica L)were obtained from the data of transcriptome sequencing.There were two or three variations in every class gene,therefore,29 CONSTANS homologous genes from mango in total were obtained.These genes were named as MiCOL?MiCOL1A?MiCOL1B?MiCOL2A?MiCOL2B?MiCOL3A?MiCOL3B?MiCOL4A?MiCOL4B?MiCOL4C?MiCOL5AB?MiCOL5C?MiCOL6?MiCOL7A?MiCOL7B?MiCOL8A?MiCOL8B?MiCOL9A?MiCOL9B?MiCOL10A?MiCOL10B?MiCOL12?MiCOL13A?MiCOL13B?MiCOL14A?MiCOL14B?MiCOL15A?MiCOL15B respectively?Sequence analysis indicated that a large difference existed among different MiCOL genes.There were 17 CONSTANS homologous genes in the modal plant Arabidopsis thaliana.According to the conserved domain of CONATANS proteins,these CONSTANS homologous genes were divided into three groups.The proteins sequence of CONSTANS homologous genes family from Arabidopsis thaliana and soybean(Glycine max(Linn.)Merr)were used into build phylogenetic tree according to the neighbor-joining method combination with MiCOL genes family.The result of phylogenetic tree showed that MiCOL genes also were divided into three groups.Group?:MiCOL?MiCOL2?MiCOL5;Group?:MiCOL1?MiCOL3?MiCOL6?MiCOL7;Group?:MiCOL4?MiCOL8-10?MiCOL12-15.The prediction results of the conserved domain in MiCOL proteins by the onlin software CD-research indicated that some special conditions existed in the structures of MiCOL proteins.Because of the special structures of MiCOL proteins,the last group were divided into three small goroups.The specific conditions as follows:In group ?:the genes only contained a CCT motif in the C-terminal but no B-box domains;In group ?a:this gene only had the secondary changed structure zick finger domain and the CCT motif but no the nomal B-box domain.It was MiCOL12 gene.In group?b:the genes contained one B-box domain and another zinc finger domain which has changed in the secondary structure in the N-terminal a CCT motif in the C-terminal.They were MiCOL9,MiCOL10?MiCOL13?MiCOL14?MiCOL15 genes;The last,In group ?c:the genes only had the secondary changed structure zick finger domain but no the nomal B-box domain and no CCT motif They were MiCOL4 and MiCOL8 genes.In addition,the expression characters of MiCOL gene in different tissues(including the mature leaves,mature stems and fowers in ' TaiNong ' mango)as well as the diurnal expression in 'SiJi' mango were analyzed by quantitative real-time PCR respectively.All primers used in this study were designed by Primer 5.0 software.The expression in different tissues results indicated that all the MiCO genes expressed in the mature leaves and mature stems,as well as flowers.The expression details as follows:Frist,in group ?,they were 2 genes(MiCOL5A?MiCOL5C)expressed highest in the mature leaves,and 2 genes(MiCO,MiCOL2A)expressed highest in the mature stems;Second,in group ?,they were 2 genes(MiCOL1A MiCOL6)expressed highest in the mature leaves,and 3 genes(MiCOL1B,MiC03B and MiCOL7B)expressed highest in the mature stems,and another 2 genes(MiCOL3A,MiCOL7A)expressed highest in flowers;Third,in group ?a,the gene(MiCOL12)expressed highest in flowers;Fourth,in group ?b,they were 2(MiCOL13A,MiCO13B)genes expressed highest in the mature leaves,and 2 genes(MiCOL9A,MiCOL15A)expressed highest in the mature stems,and another 5 genes(MiCOL10A,MiCO10B,MiC014A,MiCO14B,MiC015C)were expressed highest in flowers;Last,in group ?c,MiCOL4C expressed highest in the mature leaves,and 2 genes(MiC08A,MiCO8B),expressed highest in the mature stems.And the the diurnal expression result showed that,all of the MiCOL genes had a regular diurnal expression expect for MiCOL3B?MiCOL7A?MiCO1L13A?MiCOL14A and MiCOL15Bgenes.The expression details liked follows:Frist,in group I,they were 3 genes(MiCO?MiCOL2A?MiCOL2B)expressed highest in 4 o'clock to 8 o'clock in the moring,anaother 3 genes(MiCO5A?MiCOL5BA?MiCOL5C)expressed highest in 11 o'clock to 14 o'clock in the noon.Second,in group ?,they were 2 genes(MiCOL1A?MiCOL3A)expressed highest in 0 o'clock in the night,anaother 3 genes(MiCO1A?MiCOL6?MiCOL7B)expressed highest in 17 o'clock to 20 o'clock in the afternoon.Third,in group ?a,the gene expressed highest in 4 o'clock to 8 o'clock in the moring.Fourth,in group ?b,they were 3 genes(MiC09B?MiCOL10A?MiCOL10B)expressed highest in 0 o'clock in the night,and 2 genes(MiCO14B?MiCOL15A)expressed highest in 17 o'clock to 20 o'clock in the afternoon.Last,in group ?c,all of the genes(MiC04A?MiC04B?MiC04C?MiC08A?MiCO8B)expressed highest in 4 o'clock to 8 o'clock in the moring.This experiment may provides an important foundation of research in the function of MiCO gene families in mango.
