Jatropha curcas L.belongs to Euphorbiaceaeis,is a kind of energy plants have attracted much attention in recent years.In order not to compete with food crops and improve the ecological environment,J.curcas is generally planted in the barren and arid land,face multiple stresses,greatly affects its yield,oil quality and limits its geographical distribution,so it is of great significance to study its resistance.This study cloned 5 genes related to its abiotic stress and growth from mature leaves of J.curcas,we did bioinformatics analysis and also studied their expression patterns in different organs and under different abiotic stress,to provide reference for mining and the use of excellent resistant genes of J.curcas.The main results were as follows:1.JcNF-YB3 and JcNF-YB10 were isolated from J.curcas.The opening reading frame(ORF)of JcNF-YB3 was 549 bp encoded 182 amino acids.The ORF of JcNF-YB10 was 528 bp encoded 175 amino acids.Both JcNF-YB3 and JcNF-YB10 had the highest identity and the closest relationship with NF-YB family of V.fordii.JcNF-YB3 and JcNF-YB10 expression significantly in different organisms,JcNF-YB3 had the highest transcript profile in root,JcNF-YB10 had the highest transcript profile in seed,expression level of JcNF-YB10 was a lot higher than that of JcNF-YB 3 during seed growth periods.The expression level of JcNF-YB 3 and JcNF-YB10 were both induced by NaCl,PEG,low temperature and abscisic acid(ABA)and those treatments upregulated JcNF-YB3 and JcNF-YB10 gene expression.2.A complete CDS sequence of JcGSK was cloned from J.curcas.JcGSK contained an ORF of 1 230 bp,and encoded 409 amino acids.JcGSK had both highest identity(96%)and closest relationship with H.brasiliensis.Tissue expression results showed that JcGSK expression significantly in different organisms and had the highest expression in root,then in seeds,expression level change of JcGSK in seed had the same trends with seed growth.NaCl,Abscisic acid(ABA),PEG,cold and merchanical damage upregulated JcGSK gene expression.3.A complete CDS sequence of JcYkt6 was cloned from J.curcas,the complete ORF was 600 bp encoded 199 amino acid residues.JcYkt6 had highest identity(91%)and closest relationship with Ykt6 protein of C.melo and C.sativus.JcYkt6 expression significantly in different organisms and had the highest transcript profile in seed.Expression level of JcYkt6 in seed had the same trends with seed storage protein accumulation.ABA,PEG,NaCl,4? and merchanical damage upregulated JcYkt6 gene expression.4.Jc5?-StR gene was cloned from leaves of J.curcas,ORF of Jc5?-StR was 1 230 bp encoded 409 amino acids.Jc5P-StR had both highest identity(87%)and closest relationships with R.communis.Jc5(3-StR was one of SDRs.Jc5?-StR expression significantly in different organisms and had the highest transcript profile in seed.NaCl,PEG,4? and merchanical damage upregulated Jc5?-StR expression.In a word,JcNF-YB3,JcNF-YB10,JcGSK,JcYkt6 and Jc5?-StR genes all has tissue specific expression,may be involved in the regulation of plant growth of J.curcas although the expression patterns were different under different abiotic stress conditions,but they were able to participate in Jatropha for abiotic stress response,suggesting that they play a role in the resistance mechanism of J.curcas.
