Henan Xinyang Dabie Mountain is one of the most important distribution areas of Cuora flavomargina.However,due to habitat destruction,environmental pollution and indiscriminate killing and other reasons,the amount of wild resources of C.flavomarginata has dropped sharply.CITES is listed as rare and endangered species,endangered.In order to provide the theoretical basis for C.flavomarginata at the molecular level,this study studied the development of the microsatellite marker and the single nucleotide polymorphism marker for C.flavomarginata.And non-invasive DNA extraction was carried out with feces.The microsatellite markers and mitochondrial markers were used to study the populations of C.flavomarginata.Based on the analysis of genetic diversity and genetic structure of the populations of C.flavomarginata,the genetic differences between the populations were measured and the heterosis was predicted to provide a theoretical basis for the construction of the germplasm resource base.The protection of resources and the development of artificial breeding.The main results of this paper are:1.In this study,FIASCO method was used to construct the library of C.flavomarginata.136 microsatellite primers were successfully isolated and the polymorphic microsatellite loci were evaluated,44 pairs of polymorphic microsatellite primers were detected.2.The potential SNP loci of the genome of C.flavomarginata turtle in the endangered animals were developed by magnetic bead enrichment,and 10 primers were identified to confirm the SNP locus.3.The non-invasive method was used to extract the genomic DNA from the shell of C.flavomarginata,and it was confirmed that the extraction of DNA from the feces was not feasible,and the eggshell In the extraction of DNA is feasible,for the follow-up C.flavomarginata genetic research materials.4.Using the newly developed microsatellite markers,the genetic structure of 43 C.flavomarginata populations from three different sources was analyzed.It was found that some individuals in the farms had gene exchange between different subgroups,and the ambulance center group And the faculty group merged into one subgroup,and the two groups of the farms were treated as a subgroup.5.The mitochondrial gene CO1 and CO2 were used to analyze the haplotype6.And phylogenetic analysis of the three populations of C.flavomarginata(21)from Xinyang,and the microsatellite markers were used to analyze all individuals into Xinyang population,Anhui population and unknown population,and it is inferred that the population of the unknown area may be distant from the other than the Dabie Mountains,and the evolution relationship between the population of Xinyang and Anhui population is more near.
