Cryopreservation is widely used in scientific research,clinical medicine and human livelihood applications.In order to avoid cell damage during cryopreservation,it is often necessary to add cryoprotective agents to cell suspension before cooling.After rewarming,it is necessary to separate and remove the cryoprotective agents to avoid the side effects of the cryoprotective agents on the cells.In addition,cell therapy and other fields often need to separate and remove excess water or other substances in the cell suspension.The existing centrifugal separation method has large stress damage,cumbersome operation,and the open process is easy to cause pollution and other issues.Based on the membrane separation principle,a novel cell suspension separation and replacement system is studied.The specific contents include:(1)Based on the principle of membrane separation and separation,the theoretical model of the system is established by using the principle of transmembrane mass transfer.Based on the simulation,the simulation results show that the system can realize fast,smooth removal of cryoprotective agents in cell suspensions.(2)Based on the overall scheme and simulation results,the detailed design of the functional prototype is carried out,the key components such as the swinging device and the vibration device are developed,the control platform based on NI cDAQ is built and the control system software is designed by Labview,integrated control of the prototype.Finally,according to the industrial needs of the product,the appearance of the prototype structure,electrical wiring,etc.are upgraded,initially reached the application of medical equipment needs.(3)A number of test experiments are conducted to examine the performance of the main functional components;a series of simulations are conducted based on cell-free solutions to assess the efficiency and effectiveness of the system for cryoprotective agents treatment.The results show that the main functional components meet the design requirements,the system can be completed within 60 min 1 unit(200ml)cell suspension of the fine lines of protective agent to remove the work,and the process of cell suspension osmotic pressure continuous smooth,relatively centrifugal Law has a more obvious advantage.In this paper,through the study of membrane separation and replacement system,the design and development of human-computer interaction friendly,processing process control accurate function prototype,the prototype better overcome the current centrifugal processing equipment some important drawbacks,such as cell damage caused by high centrifugal force,cell pollution caused by the open process.It is expected to cryopreservation,cell therapy and other industries play an important role.
