Expression,Purification And Crystallization Of Mugwort Pollen Allergy-Associated MHC Proteins

Allergic disease is a common clinically diseases.Pollen is one of the most important allergens.The sensitized pollen from allergizaion plants floating in the air,causing a series kinds of allergic diseases.And ths in year,the rate of pollen allergy is rising year by year,following with season.Mugwort,belong to artemisia and compositae,are distributed in Asia and Europe and widely distributed in China.Wherein Artemisia pollen is more complex for the allergenicity.A variety of pollen allergens have some cross allergenicity.As the high ability of adaptability and prolificacy,the wild resources mugwort are widely distributed and expanding.And because of its' high environmental adaptability and strong ability to reproduce,wild resources are widely distributed and expanding.So,it is urgent to know the mechanism of mugwort pollen allergic.The mechanism of mugwort pollen allergy is reported as: mugwort pollen allergen proteins hydrolysis to peptides,which is captured by MHC(major histocompatibility complex,short for MHC,also called HLA in humans)of cell surface in vivo.The MHC presents peptide antigens to the TCR(T cell surface receptor).And formed the complex of TCR / MHC / peptide.This complex activates the T cell.The activated T cell kill cells with the MHC / polypeptide,and leading to allergy eventually.This paper synthesized related MHC molecules of mugwort pollen allergy proteins-DR1*01:01,DR1*04:01's gene.And then overexpressed in E.Coli.,while DR1*01:01 express normally in inclusion bodies,but DR1*04:01didn't express.So we only got the protein of DR1*01:01 by refolding.Then purified by multi-step,multi-strategy method.Finally,we got a good trait complex.Through screening many crystals kits,we got preli minary crystal.After that we will optimize deeper screening to obtain the information of composite structure.It provides a theoretical basis of subsequent drug design and allergy prevention.
