Dopa is a biologically active substance. It is both the body's neurotransmitters and also a neuromodulator in the central nervous system and peripheral organs with varying degrees of regulation. Dopa can be divided levodopa and dextrodopa according to their chiral structure. The study found it can be converted in the body to 35 substances. Catecholamine has been concerned. The early 21 st century, its research results have won two Nobel Prizes(Nobel Prize in Physiology or Medicine and Chemistry Award). Recent studies have found that Dopa and Conversion substance plays an important role in the nerve- immune system.This paper aims to study metabolism network-Dopa in vivo, as levodopa convers to 3,4-dihydroxyphenyl lactic acid. According to physiological and biochemical properties of the compounds, the relationship and physiological significance is analyze between dopa and 3,4-dihydroxyphenyl lactic acid into, providing to a potential reference value for the central nervous cardiovascular and cerebrovascular diseases. The results as follows:1. To establish an HPLC conditions and detect to dopa,3,4-dihydroxyphenyl pyruvic acid and 3,4-dihydroxyphenyl lactic acid of biological samples. Selection HPLC- electrochemical detection with Agilent 5 HC-C18(250mm × 4.6mm, 5? m) column, was used with methanol-1m L·L-1-carboxylic acid(p H was adjusted to 4.0)(3:97) as the mobile phase, isocratic elution, the flow rate of 0.6m L·min-1, the column temperature was 30 ?, the detection voltage 0.9m V, detection current 50 n A.2. Using Oasis HLB and BCX SPE column. Tthe use of solid phase extraction techniques for biological sample pretreatment. Levodopa, dopamine, 3,4-dihydroxyphenyl pyruvic acid and 3,4-dihydroxybenzene average recoveries were 82%, 91%, 80% and 84%.3. Detection of biological samples dopa conversion substance. Healthy SD rats was administrate L-Dopa(?9 day, ?200mg / kg), and can detect 3,4-dihydroxyphenyl lactic acid and 3,4-dihydroxyphenyl pyruvic acid in the urine. Levodopa can be confirmed to the conversion of 3,4-dihydroxyphenyl lactic acid in the body. 4. Analysis of Dopa and chiral structure in biological samples, respectively use GC/MS and HPLC. The peak of 3,4-dihydroxyphenyl lactic acid is poor, R <1min.All of all, this study suggests that a high intake of dopa can cause physiological changes in the body, and this physiological changes may promote the conversion of DOPA to DSS, in which the transformation process of the body spontaneously secrete endogenous substances 3, 4-hydroxy acid. and it has a similar chemical structure with DSS. The prime years, DSS is against to inflammatory, kill normal flora, so that the body heal itself or symptom relief efficacy. The immune system to dynamically adjust the self-acting, maintain the physiological balance.The conversion is not only beneficial demonstration the material relationship of traditional Chinese culture, "Man and Nature harmonization", but also a powerful interpretation of immunology "in vivo changes in the material self-adjustment". A study provides a new idea from danshensu convers to dopa.
