Saline soil is important land resources in China,Salt stress is one of the major abiotic stress in our country,it can affect the normal physiological activities and crop yields., it has significant growth potential. Halophytes is common natural flora on saline soils, they have important economic values. Exploring the salt tolerance mechanism of halophytes and screening salt-tolerant genes have become more economic and effective measures.Halogeton glomeratus is one of annual dicotyledonous plants belong to Chenopodiaceae. They mainly distributed in desert and saline-alkali land in gansu, qinghai province etc. Physiological studies have shown that they have strong salt resistance. After salt stress, vacuole in mesophyll cell can accumulate a large amount of sodium ions. Using the technology of molecular biology to find out Halogeton glomeratus' s Salt-tolerance mechanism and Salt-tolerance gene.It has great significance to cultivate salt-tolerant varieties.On the basis of transcriptome sequencing on Halogeton glomeratus after salt stress. Screened four gene which have up-regulated expression. Using RT-PCR and RACE to get the full-length of gene,This main resullts of this experiment are as follows:1.After 3'RACE,we have got three sequences:Unigene22136,Unigene33874 a-nd Unigene34165. After joining together, the sequence of Unigene22136 is 609 bp, the sequ-ence of Unigene33874 is 471 bp, the sequence of Unigene34165 is 760 bp,2.After 5'RACE and 3'RACE,we have got a full-length sequence:CL5227, the sequence of it is 501 p,and encoded for a polypeptide of 41 amino acids with an molecular weight of 4.56 k D,Through bioinformatics analysis,The Cl5227 protein belongs to hydrophobin and basic protein.the protein has no signal peptide and transmembrane domain. Located in the mitochondria, with functions of growth factors, structural protein.3.we have cloned CL5227 from Halogeton glomeratus and built a plant expression vector: p CAMBIA3300-CL5227. Imported into agrobacterium successfully and Impregnated arabidopsis.Now we have got T1 generation plants. Laid the foundation for further identification of gene function.
