Lgl was firstly described as tumor suppressor proteins in Drosophila,and it's very conserved in C.elegans,Drosophila, Mammalia and human. In many cancer cases, Hugl-1(human Lgl1) is found decreased expression or no expression. But the role of Hugl-1 in development of certain cancer(e.g. liver cancer) is still unclear.For the reason of ethics and the lack of experimental material from human, we can't directly knock-out gene on human. But fortunately, we can knock-out gene in mouse. This protocol reference to the several international common gene conditional knock-out strategy, and we successfully obtained mice that specifically knock-out Llgl1 in liver. The identification method in this protocol is simple and suitable for high throughput work. The conditional knout-out vector can be simply modified for another gene or preparation of Homozygous knock-out ES cells and mouse either. |
