Research And Characterization Of Non-Coding Function Of GAB2 Gene

Objectives:Constructing 3 types of plasmids which contain CDS, FL and FLM of Gab2 gene, find if Gab2 gene has some non-coding functionsMethods:Constructing 3 types of plasmids which contain CDS, and the transient transfection breast cancer cell line and colon cell line. Using Western Blot to test the expression of the protein in the transient cell line. Besides, we also do some cell function experiments and Q-PCR testing, so we can identify the differences between CDS and the FL, then speculating if Gab2 gene has some non-coding functions besides coding functions.Result:We constructed the 3 types of plasmids which contain CDS, FL and FLM of Gab2 gene, and the transient transfection cancer cell lines. Many cell function experiments and most Q-PCR testing showed that the CDS was little similar with the FL and even there were some contraries between CDS and FL.Conclusion:The function of Gab2 gene may not just depend on the CDS of it. The non-coding sequence of it may have very important function so that making huge differences between CDS and FL. To research a gene, even a familiar coding gene, we should observe the full-length but not just the CDS.
