DEAD-box helicase protein family are ubiquitous,highly conserved enzymes.One of the motifs(Asp-Glu-Ala-Asp amino acid sequence)gives the protein family its name.The family is classified into two kinds of proteins:DEAD-box RNA helicase and DEAD-box DNA helicase.As an important member of DEAD-box RNA helicase protein family,DDX3X utilizes ATP to bind or remodel RNA and ribonucleoprotein complexes as it has been widely investigated.DDX3X plays important roles in many intracellular pathways,such as mRNA transcription regulation,mRNA nuclear export,cell cycle progression,apoptosis,cancer,immune response and so on.Many researches have been focusing on the function of DDX3X,however,that DDX3X plays a role in microRNA and other small RNAs pathway is still unclear.In this paper,on one hand,we found that microRNA has some specific change when DDX3X was knocked down.On the other hand,DDX3X can affect the chromosome separation.To further investigate the function of DDX3X,we used the CRISPR/cas-9 technology to establish the knockout stable cell line.
