Miss Gao Youfei,a colleague of mine,had gotten a novel MYB transcription factor Gm MYB173 in soybean Zigongdongdou through the yeast one-hybrid system.To investigate the basic function of GmMYB173,the RT-PCR of Gm MYB173 in different tissues,the subcellular location and the transgenic function analysis assays were done.Exploring the function of GmMYB173 in soybean flowering regulation and its function on the soybean development would contribute to the soybean breeding research.The main study methods and results were as follows:1.The amino acid analysis of GmMYB173: Gm MYB173 contained one MYB domain and belonged to the R1/R2-MYB transcription factor.The prediction of subcellular of GmMYB173 was in the nucleus and without signal peptide.The amino acid sequence of GmMYB173 contained binding polypeptides of 14-3-3 protein.2.The results from particle bombardment assay using onion epidermal cells and PEG/CaCl2-mediaed transformation of Arabidopsis protoplasts suggested that GmMYB173 protein mainly expressed in the nucleus,which was in keeping with the bioinformatics prediction.Gm MYB173 protein without 14-3-3 binding polypeptides lost the nuclear-specific feature.These results suggested that 14-3-3protein would influence the subcellular location of GmMYB173 and further regulate the function of Gm MYB173.3.GmMYB173 could interact with 16 kinds of 14-3-3 proteins that were examined in soybean through the yeast two-hybrid assay.However,the?-galactosidase activity of interaction was different;the interaction of Gm MYB173 and GmSGF14n displayed the strongest.These results suggested that GmSGF14 n might interact with GmMYB173 and then regulated the function of target gene in plant and GmMYB173 function was regulated by different 14-3-3 protein.4.The relative expression level of GmMYB173 in leaf and epicotyl was higher and it could response to the ABA and drought stress through the RT-PCR,which was consistent with expression pattern of potential target gene GmFT2 a.The transgenichair-roots could live better in the medium with 5% D-mannitol medium than the control;this suggested that GmMYB173 could response to adversity stress.5.Heterologous expression of GmMYB173 in Arabidopsis slightly promoted the early flowering,while the expression of FT and SOC1 were up-regulated.The over-expression of GmMYB173 in soybean hair-roots accelerated the expression of GmFT2 a and GmFT5 a the key genes in the regulation of flowering,these could promote the early flowering of soybean.T1 generation of GmMYB173 transgenic soybean was gotten by the Agrobacterium-mediated transformation,and through T1 phenotype observation,we hypothesised that GmMYB173 regulated the soybean flowering by shortening the vegetative growth stage.
