DNA is the carrier of genetic information,there are many types of DNA damage,the most serious one is DNA double-strand breaks(DSBs),if can not be repaired,cell cycle will be stalled,or sometimes,lead to apoptosis.DNA of Eukaryotic cell must be packaged into chromatin,it can block gene's expression,replication and repair.To improve the accessibility of these parts of DNA,cell must loose the structure of chromatin,so the effector can come close to it.In the process of eukaryotic DNA damage repair,chromatin remodeling is of great importance.Different chromatin remodelers can affect the interaction between DNA and histones,then improve certain parts of DNA's accessibility.Lots of chromatin remodelers has been deeply researched,now we focus on the chromatin remodeling factor CHD4,it's an ATP-dependent chromatin remodeling complex,some experiments show that CHD4 take part in ionizing radiation mediated double-strand break repair.This paper mainly discussed the role of chromatin remodeler CHD4 in ETO mediated DSBs.Though western blotting,and RNA interference we found that CHD4 truely involve in this process.Immunoprecipitation experiments indicated that CHD4 did not interact directly with ?-H2AX.P300 and CHD4 interacted directly with each other.This experiment provides a reference for further study of CHD4 in DNA double-strand breaks,it also provides new clues for the understanding of chromatin remodeling mechanism in DNA double-strand breaks repair process.
