In this dissertation, we have carried out some original studies including fingerprint spectrum, metal element content, protein category of Hainan Alpinia Katsumadai and the biological activity of its main active components.The thesis consists of six chapters.Chapter 1:The study progress of Alpinia Kastumadai Hayata was summarized. Its main active components and their biological activity were also discussed in detail both at home and aboard.Chapter 2:The fingerprints of Hainan Alpinia Katsumadai and analytical method determining its main active components contents have been established by different spectroscopic methods. Fourier transform infrared spectroscopy(FT-IR), UV spectroscopy and fluorescence spectroscopy were employed to analyze Alpinia Katsumadai harvested from Hainan Baisha and Bawangling. The results indicated that both ethanol extracts of Alpinia katsumadai Hayata harvestd from Baisha and Bawangling have the similar spectral signatures. And it also revealed that alpinetin and cardamomin are two principal active components of Alpinia Katsumadai.However, there are obvious differences in light of second derivative spectra, which was apparent to identify and distinguish Hainan Alpinia Katsumadai from different geographical regions. An UV-quantitative analysis method with good precision and recovery for alpinetin and cardamonin in Alpinia Katsumadai was established according to the relevant absorption principle.Chapter 3:Seventeen kinds of metal elements of Hainan Alpinia Katsumadai including potassium, calcium, magnesium, sodium, aluminum,ferrum, manganese,copper, Zinc et al were determined by using microwave digestion- inductively coupled plasma mass spectrometry(ICP-MS). The results showed that the content of metal elements such as potassium?calcium?magnesium and aluminum was higher and potassium had highest content.Chapter 4:The interaction between alpinetin or cardamomin and blood protein was investigated by proteomic analysis. Under simulated physiological conditions, 10 differential displayed protein spots were identified by using two dimensional gelelectrophoresis( 2D-electrophoresis) associated with Matrix assisted laser desorption time-of-flight mass spectrometry(MALDI-TOF-MS).Chapter 5:The protein categories of Hainan Alpinia Katsumadai from Hainan Baisha and Bawangling were identified. The optimized 2D-electrophoresis protocol combined with MALDI-TOF-MS was successfully applied in protein analysis of Hainan Alpinia Katsumadai from different geographical regions. There were 40 differential displayed protein spots in initial verification.Chapter 6:The major conclusion and perspective carried out in this dissertation were summarized.
