Long non-coding RNA(LncRNA) is a RNA sequence that is more than 200 nucleotides in length which does not encode protein. LncRNA can regulate the expression of genes at the level of transcription, post transcriptional and translation.LncRNA has development stage and cell type specific temporal and spatial expression model, indicating that it plays an important role in the process of organism activities.In the previous work, we found a new long non-coding RNA Lnc784-an antisense RNA of TCF7l2. TCF7l2 is an important transcription factor in the Wnt signaling pathway, which plays an important role in the development and maturation of the oligodendrocytes. In addition, mutations in the TCF7l2 gene increase the risk of developing multiple sclerosis and type two diabetes. Therefore, it is of great significance to study the role of Lnc784 in the regulation of TCF7l2 gene expression.In this article, we analyze the expression of Lnc784 and TCF7l2 in different neuroanatomical regions of mouse brain and they have showed obvious differences in different neuroanatomical regions of mouse brain, both in the cortex expression were the highest, the olfactory bulb were the lowest. When up-regulated or down-regulated the expression of Lnc784 in mouse adult neural stem cells, the results of q-PCR and Western blot test indicate that Lnc784 plays a negative role in the regulation of TCF7l2,expression in both transcription and translation levels. The effect of up regulation or down regulation of Lnc784 on the expression profile of neural stem cells is analyzed by high-throughput depth sequencing, the results confirmed that Lnc784 is negatively related to the expression of TCF7l2, and it is found that Lnc784 plays an important role in the differentiation of glial cells, insulin secretion and glucose metabolism. The results of this study may provide a new way for the treatment of multiple sclerosis and diabetes.
