Aquilegia viridiflora belongs to the family Ranunculaceae,Aquilegia includes about70 species,which are widely distributed in the northern hemisphere temperate regions.In China,about 13 species in are found,of which,the species A.viridiflora is a widely distributed across northeast and north of China.In this study,we selected seven natural populations of A.viridiflora,which show differences in flower morphologies and colors.To evaluate the genetic structure,we sequenced three single copy nuclear(ADH,OGGA and gEST8)and ten(trnD-psbM?mat K?ndJ-trnF?ndhC-trnV?Ycf6-psbM?rps12-rp120?atpH-atpL?ndhA?trnC-ycf6 and,trnC-rpoB)chloroplast genes of these populations.Based on the obtained genetic data,we conducted genetic structure,haplotype network and phylogenetic analyses.The nucleotide diversity results show that the species A.viridiflora possess high genetic diversity.In addition,we also found that the haplotype network did not reveal geographic distribution pattern.Based on the genetic differentiation analysis,our results showed that the species shows obviously intra-specific genetic differentiation.Taken together,our results demonstrated that both morphological and genetic differentiation occurred within this species.We therefore proposed that the genetic drift under geographic isolation and natural selection have together shaped the genetic structure of this species.
