Gamma-aminobutyric acid(GABA) is the main inhibitory neurotransmitter in the central nervous system, it exert its biological function by GABA receptor. There are two classes of GABA receptors in nerve cells, respectively is ionic gamma-aminobutyric acid receptor-GABAA receptor(GABAAR) ? GABAC receptor(GABACR) and metabolic gamma-aminobutyric acid receptor- GABAB receptor(GABABR), GABABR belong to the G protein coupled receptor(GPCR) C family, it can be mediated slow and long process inhibiting effect of synaptic transmission. Located both pre-synaptically and post-synaptically, GABABR is thought to play a role in CNS disorders such as epilepsy,spasticity, schizophrenia, anxiety, depression, cognitive deficits, and addiction. Therefore,GABABR became the treatment drug targets of a variety of nervous system disease and mental disease.Drosophila melanogaster, commonly known as "Fruit fly", is an ideal model organismin in the experiment of genetics and developmental biology, in recent years, it,s being increasingly used as a model organism in neurosciences, including neuropharmacology.Because Drosophila melanogaster compared with the typical mammalian model has many advantages, such as using the Drosophila melanogaster we can obtain better operability of the genetic, under lower cost can get countless subjects, and fewer regulatory restrictions.Using Drosophila as a model, it has been shown that GABABR are involved in the behavioral actions of alcohol and g-hydroxybutyric acid, and possibly in pain.Furthermore, recent research suggests that in flies these receptors may play an important developmental role and that they participate in olfaction and in regulation of circadian rhythms.However, we still should recognize that the essence of the difference between insects and mammals, only more clear understanding of Drosophila GABAB receptor(DGABABR)and the difference between mammals GABAB receptors and DGABABR,also the cause of this difference, we can better use the Drosophila model instead of mammalian model to do research in neuroscience, including neuropharmacology.In this study, we firstly studied the expression patterns of DGABABR in membrane,and were compared to the DGABABR membrane expression patterns with mammals GABAB receptors membrane expression patterns to find out the differences between them.Then we studied what is the reason of these differences, we found that L777 amino acid residues located in DGB2 C terminal play an important role in interactions of DGB1 C terminal between DB2 C terminal.In conclusion, this research is mainly studied the interaction between the C terminal of DGB1 and DGB2.we found the important areas of DGB2 C terminal in the interaction between the C terminal of two subunits in DGABABR, and extends 2001 Mario Mezler et.Al predicted DGB2 coiled- coil- domain to L777.
