The structure of protein is very complex, only the correct folding can complete its corresponding biological function, when the error folded proteins can not get timely removal, then the large number of accumulation will cause the Protein conformation disease. In recent years, the incidence of neurodegenerative disease caused by the misfolding protein is higher and higher, makes the study on protein folding problem become very popular. Because of the complexity of the problem of protein folding, the simple tag observation ways and some algorithms based on the research of protein folding mechanism cannot satisfy the requirement of protein detection, then more rapid, nondestructive and high sensitivity detection method is in urgent need. In this paper, according to the protein thermal condensation problem, put ting forward a detection method based on the phase modulation SPR imaging,drawing the curve of the reflection phase information changes caused by biological molecules interact with each other, calculating the experimental results that we need. In order to reach the goal of the above, this paper mainly completed the following work:Establish the experiment platform of protein thermal condensation detection based on phase modulation SPR imaging, built the mathematical model of the same molecular interaction. We get the conclusion through the mathematical deduction: aggregation degree of lysozyme molecules is not obvious at the beginning of the heated, but the aggregation degree becomes strongly after reaching a certain temperature, gathering process achieve dynamic balance after reaching maximum temperature. Mathematical deduction is consistent with the experimental results.Use the algorithm of median filtering and wiener filtering to analyze the images which get in this paper. We get the concluded based on the MATLAB simulation results that: wiener filtering algorithm can better restore image pixel information. This article uses the wiener filtering algorithm to deal with image noise which produced during the process of obtaining, to be prepared for the subsequent phase information extraction.Introduce the methods of the extracting of image phase information by interferometry, and comparing the existing algorithms for this article test environment. Analyze every algorithm's ability of resisting the noise and the size of the error. Eventually choose the algorithm of Stoilov for phase information extraction algorithm of this paper.Verify the performance of the device through the test, and the device is applied to detecting the lysozyme thermal condensation, fitting the real-time detection signal curve of the lysozyme thermal condensation which got through the test. Finally we got the kinetic parameters and the equilibrium constant of the lysozyme thermal condensation.Through the above work come to a conclusion that: phase modulation SPR method is very suitable for the study of the lysozyme thermal condensation. Compared with other methods, has the advantage of higher sensitivity, rapid and real-time detection, not using of the tag sample, test procedure is simple.
