Emei Mustache toads (Vibrissaphora boringii) belongs toFamily of Megophrys Anura Vibrissaphora genera, which isdistributed in Sichuan, Hunan, Guizhou, Yunnan and Guangxiprovinces.The complex terrain of distribution area is blockinggene flow between different geographical populations, leadingto significant genetic differentiation among populations. Totest this hypothesis and explore the genetic differentiationreason of two isolated Emei Mustache toads populations, thisstudy analyzed the genetic variation condition of two isolatedEmei Mustache toads populations according to mitochondrialDNA molecular marker and the significant geological changestime. This study analyzed the of genetic variation andpopulation genetic differentiation pattern from76samples ofSichuan and Hunan two isolated populations applicationmitochondrial DNA Cytb gene sequence analysiThe results are asfollows:1. There have higher genetic diversity in differentgeographic populations of Emei Mustache toads (Vibrissaphoraboringii). There exist67variable sites of1097bp sequencein33individuals of Hunan geographic populations and defined13haplotypes. The haplotype diversity (h) was0.814, nucleotide diversity (?) was0.029. There exist4variablesites of1097bp sequence in43individuals of Sichuangeographic populations and defined8haplotypes. The haplotypediversity (h) was0.671, nucleotide diversity (?) was0.001.There were90variable sites and21haplotypes of76individuals of two geographical populations. The haplotypediversity (h) was0.861, nucleotide diversity (?) was0.030.2. There are not shared haplotype of two populations. Thegenetic differentiation Fst was0.73(P <0.05) significantlygreater than0.25between the two isolated populations, whichindicate that the gene flow is relatively poor betweenpopulations, genetic differentiation is significantly.3. The results show that it can be can be arranged into therehighly supported groups?branch A?B1and B2according to thePhylogenetic tree analysis(MP method and Bayes method).4. The divergence time of branch A and branch B areestimated to diverge at4.45-10.17Ma BP, according to theof molecular clock theory. It may be caused by Quaternary iceage. The Quaternary glacial affected the gene flow of differentgeographical populations and produced a significant geneticdifferentiation.5. Using of Arlequin mismatch distribution and Tajima's neutrality test analysis found: Sichuan populations undergosignificant population expansion, and Hunan geographicpopulations remained stable branch.
