Microscopy is essential in medical and biological research, however, the available observe methods are simple than only the slice of the sample can be observed instead of the stereo chemical structure due to the shallow depth of field of the system. The available microscope can’t meet the demand of scientific research since only one 2-dimensional picture can be acquired at a time and the sample is easily destroyed while it is need to be flipped. A kind of light field spectrometry microscope imaging system was designed with the core device of microlens array based on the combination of the theory of light field and the dispersion theory of the spectral splitting system. It can acquire the light field and spectral information of the object in one exposure time.Microlens array was set at the image plane of microscope objective in the system. Different perspective information and deep depth of field of the object can be obtained when microscope objective is combined with multi-channel simultaneous imaging microlens array. Every single lens image different part of the specimen, high resolution image can be reconstructed from the sub image array obtained by detector when the reconstruction algorithm is used to deal with the image recorded on detector. LFM(light field microscope) is combined with spectral splitting system to constitute light field microscope imaging spectrometry considering the identification of the specimen, the sub image array is dispersed effectively due to the blank space of the image plane. Different wavelength is dispersed into different position according to the grating diffraction formula or prism splitting principle. Light field microscope imaging spectrometer can obtain five dimensional information of the specimen while traditional imaging spectrometer obtains three dimensional information of the specimen.Five dimensional formula was derived in this article according to the light field and spectral dispersion theory, the structure of LFM and light field spectrometry microscope imaging system was simulated through Zemax by studying the theory of both the two system, the effect on the imaging produced by the matching relation and error of each link was analyzed later, which optimized the whole imaging spectrometry system. In addition, good imaging quality was obtained with optimized reconstruction algorithm by combining light field microscope algorithm with interpolation and filter algorithm. In the later stage, experimental platform was set up to verify the simulation results.
