Three dimensional phase microscopy tomographic(TPM) is a new type of microscope system which is developed on the basis of quantitative phase microscopy(QPM) of the refractive index distribution of biological samples in vivo measurement. TPM is reconstructed by phase projection and multi-angle rotation, and the three-dimensional refractive index distribution of biological samples is reconstructed by a certain algorithm.The choice of the reconstruction algorithm is an important factor affecting the spatial resolution and the accuracy of the refractive index.With the increase of the number of cells in the sample and the sample structure becoming more complex, the diffraction of light wave will become very obvious, and the quality of the image will be greatly affected. Therefore, we must use better adaptability reconstructionalgorithm for the effects of diffraction of light.In this paper, based on the above problems, we introduced the first order Born approximation and the Rytov approximation in the image reconstruction, and introduced the hybrid approximation of the first order Born approximation and Rytov approximation. By adjusting the power of the two approximation to maximize the advantages of the two reconstruction algorithms, the accuracy of image reconstruction is improved. The main contents and contributions of this paper are as follows:1. In order to verify the validity of the hybrid approximate algorithm to reconstruct the complex sample, we carried out the simulation experiment and synthesized three-dimensional correction Shepp-Logan S-L model of scattered field data to provide a quantitative basis for the image processing algorithm.2. In order to further discuss the adaptability of the hybrid approximation to the real experimental data, the real experimental data of a single polystyrene bead sample and a single layer of polystyrene beads were respectively reconstructed, and the reconstruction results were evaluated.
