Biosensor technology is an interdisciplinary technology, the electrochemical impedance spectroscopy detection based on aptamer biosensor is widely used in analysisingkineticof electrode and quantitative detection,which is highly sensitive、low cost and easily miniaturized.Aptamer biosensors show their characters whenthe detecting environment change. Beside the influence of aptamer biosensor,the system building、condition setting and the data analysis also resulting in the change of detection result. In this paper,we try to analysis the impact factors of the detection, details are as follows:1. Make a deep understand of the electrochemical impedance spectroscopy detectionwhich based on aptamer biosensorand make clear the principle、basic conditionand error source of the detection.2. Completing the detection in the 3-electrode system and processing data onMATLAB to get the kinetic parameters.3. Discuss how the basic voltage influence the aptamer biosensor in structure、performance and specificity.4. Describe the equivalent circuit with and without using the Luggin,and whether theLuggin is necessary in detection.5. According to the analysis of the detecting factors,a suitable condition is selectedfor the detection of cadmium with aptamer bioseneor by electrochemicalimpedance spectroscopy. The result shows that the change rate of electron transferresistance ?R% against the logarithm of cadmium concentration is linear overthe range from 1nM to 40 nM,which is lower than the detecting limit reported inthe article of the aptamer selection.6. Evaluate the aptamer biosensor in real sample.
