Studying of the theory of Hollow-core Metal-cladding waveguide(HCMW), which has high sensitivity has been focused. According analyzing the theory of HCMW, can find the waveguide as bio-chemical sensor has obvious advantages. That’s because the Ultra-high order modes(UOM) can be excited on the HCMW. There are two factors: 1.The coupling into the energy of the light field can be focused on the wave guide layer of HCMW; 2.There are the refractive index and structural material factors. Apparently waveguide has little effective refractive index, leading to high sensitive to the change of environment and very narrow ultra-high derivative absorption peak can be clearly shows the change of environmental factors in the waveguide. These because of the existence of oscillating field conditions made hollow metal cladding waveguide as the sensor sensitivity is much higher than the average of the evanescent field sensor.In this article two applications on concentration detecting and the displacement measuring of this sensor was mentioned. We have make analysis on the sensitivities of sensors in our experiment. And make the simulation on the ATR spectrum of the waveguide used in the experiment. As the imaginary part of the refractivity index of samples mainly affect the up-and-down movement of the ATR spectrum, the method which detect the imaginary part of the refractivity index have high sensitivity.This paper, Hollow-core metal-cladding waveguide building from the whole angle of scanning optical oscillating field sensor of a complete set of experimental system, and use the sensor to measure some samples in the field of biochemical(including: the epidermal growth factor, endosulfan and chlorella).Measurement experiment of these samples, is by changing the concentration of the sample under test or the proportion, make the ATR spectra of change. The experimental results show that the sensor can distinguish the tendency for 0.111 mm/ L of the change of the concentration of chlorella, far better than chromatography is used to measure the results, the sensor can analyze the different structure of endosulfan and the ratio of two structures can be analyzed, the sensor can also analyze the epidermal growth factor and drug interaction time and dosage of control process, etc.
