Chemiluminescence (CL) sensors based on optical fiber are powerful tools for clinical medicine, biochemical research, environmental protection and food industry, owing to their small size, convenient use and fast assay. Among them, CL sensors based on enzyme is getting more and more attention. As a CL catalyst, enzyme has advantages such as high selectivity, mild reaction conditions, and free of pollution, and are often applied in this kind of sensor. But natural enzyme is not stable, and easy to inactivate, which makes it hard to reuse. Later the concept and technology of immobilized enzyme was proposed and developed. The sol-gel encapsulation method has been widely used in enzymes immobilization since it can be prepared under mild chemical conditions, allowing the entrapped enzymes retain their activity.As a promising special fiber, hollow fiber (HF) has advantages of simple structure, easy fabrication, and flexibility. The air-core feature implies that it can be used as CL reaction cell when transmitting CL signal. Transmission characteristics of CL in both Ag and SiO2 HFare studied. The results show that HF has a low loss transmitting CL signal and a good linear response to reagent concentration. Also flow rate and fiber length can be optimized to improve the system. Because reagent is corrosive to Ag, dielectric-coated metallic HF is more suitable to be used in the CL system. The thickness of dielectric layer can be optimized according to target wavelength, and also can protects Ag layer from corrosion.In this paper, a new kind of CL sensing cell based on HF is introduced by combining sol-gel enzyme immobilization with HF together. The technology of coating sol-gel sensing film inside of HF is studied. Transmission mode of this new device is studied by theoretical analysis, and optimization of system parameters is discussed according to the calculation results. A HF based sensing system is designed and established for detecting hydrogen peroxide concentration. And it is applied to detect the hydrogen peroxide concentration in contact-lens disinfectant solution. A high sensitivity (1OμM) and good repeatability is achieved.
