Objective:Observed the expression changes of P53 signaling pathway(P53 、 Bcl-2 、 Bax 、 P21 、 Survivin) in mouse subcapsular renal transplantation tumor of H22 to prove the anti-cancer effects of the aerobic exercise and macrothele raveni venom intervention and its possible molecular mechanisms.Methods:Male KM mice were choosed to establish tumor models by transplanting H22 cell strain under the mouse renal capsule. All alive model mice were randomly divided into 5 groups(n=10): the model control group, the treadmill intervention group, the spider venom intervention group, the combination intervention group(treadmill and spider venom), the positive control group.Results:(1)Tumor inhibition rates: the combination intervention group with 64.82%, the spider venom intervention control with 45.43%, the positive control group with 24.65%, the treadmill intervention group with 9.76%.(2) HE staining: Firstly, the microscope could saw tumor tissues, which proved that the model was successful. Secondly, in the model control group, tumor area was relatively large with the tumor cells obvious atypia, significantly larger cell body, and large and deeply stained nucleuses, disorderly and densely arranging, invasively growing like nests,with less intercellular substance; in the spider venom intervention group and the combination intervention group, tumor tissue showed a relatively complete boundary,a decreased density and a loose distribution with cell shrinkage, karyopyknosis, cytoplasmic concentration and vacuolization. Treadmill intervention group was no typical apoptotic morphological changes.( 3) The Immunohistochemistry observation: Based on the sequence from the model control group, the spider venom intervention group to the combination intervention group, staining intensity of mt-P53 、 BCL-2 、 SURVIVIN respectively decreased, BAX 、 P21 increased.( 4) The Real-time PCR detection: Based on the sequence from the model control group, the spider venom intervention group to the combination intervention group, mRNA expressions of Bcl-2、Survivin respectively decreased, P53、P21 increased.Conclusion:Interventions of macrothele raven venom or aerobic exercise combined with spider venom had curative effects in H22 transplantation tumor under the mouse renal capsule. The molecular mechanism chould be to positively regulate the activity of P53 signaling pathway to inhibit tumor cell proliferation and induce tumor cell apoptosis.
