ã€Objective】: Obese is related to diabetes, cardiovascular disease, insulin resistance and other diseases, and it has become a difficult problem in the world wide. Obese result from fat accumulation partly, and many other factors involved in it, but the detail mechanism is not clear currently. Zinc Alpha2 Glycoprotein(ZAG) is a new fat mobilization factor, lipolysis is one of the most important functions of ZAG. In the study we observe effects of the intervention of exercise and ZAG to the Expression of FAS and HSL in the liver of obese mice and discuss its mechanism by gene expression. This can help to find a new way to treat obese, and proved evidence for the method of exercise and exogenous ZAG to deal with obese. ã€Methods】: The pc DNA3.1(-)-mZAG plasmid was transformed into TOP10 competent cells, and screened the recombinant which containing plasmid by the method of resistant labeling. Then amplified, extracted and purified to obtain large amounts of ZAG plasmid. The plasmid is used for transfection obese mice.140 eight-week old KM male mice, randomly select 40 mice feed normal diet, other mice feed high fat diet, to build obese model.The obese mice divide into four group : Placebo group(-EP, n=7), ZAG group(-EZ, n=9),Exercise and ZAG group(EZ, n=6), Exercise and placebo group(EP, n=7).-EP group and EP group inject 300 ul OPTI-MEM and 10 ul Lipofectamine,-EZ group and EZ group inject 300 ul OPTI-MEM and 10 ul Lipofectamine and 5ug ZAG placebo(8 weeks Intervention, inject twice a week); EP group and EP group were administered with 8 weeks moderate intensity treadmill running( 10m/min for the first four weeks, 12m/min for the last four weeks, 60 min, 5 days/week).extract the total RNA of liver, and test liver ZAGã€HSLã€FAS mRNA level by real time PCR.Analysis data with SPSS18.0 system, all the data represent with Mean±S. Two-sample mean comparison with independent samples T-test, two factors using multivariate analysis of variance. Two-tailed P <0.05 was considered statistically significant difference, P <0.01 considered very significant difference. ã€Results】: 1. Amplified, extracted and purified ZAG successfully, and used for transfection obese mice.2. Induce the obese model successfully: the weight of mice with normal food is 53.93±2.37 g, the weight of mice with high-fat food is: 67.03±4.68 g, he weight of mice with high-fat food is 1.23 times of the weight of mice with normal food, obviously we built the obese model successfully.3. The effects of exercise and ZAG to the mice’s weight ã€visceral fat mass and fat-body index: The weight trend of –EZ and EZ group are decreased, and the weight trend of –EP and EP group are increased, but the weight gain of EP group is lower than –EP group(P>0.05); the visceral fat mass and fat-body index of –EZ and EZ group are respectively lower than –EP(P>0.05) and EP(P>0.05) group.4. The effects of exercise and ZAG to the expression of ZAGã€HSL and FAS mRNA in mice liver: ZAG expression of –EZ and EZ group are higher than –EP(P>0.05) and EP(p<0.05) group respectively; ZAG expression of EP and EZ group are higher than –EP(P<0.05) and –EZ(p<0.05) group respectively; HSL expression of –EZ(P<0.05) and EP(P<0.05) group are higher than –EP group; FAS expression of –EZ(P<0.05) and EP(P<0.05) group are lower than –EP group. ã€Conclusion】: 1. High fat food increase the weight may by inhibit the expression of ZAG and HSL, increase the expression of FAS in the liver.2. Both exercise and inject ZAG plasmid can decrease fat accumulate and decrease the weight at last,it may relate to increased expression of ZAG, and then ZAG up-regulate the expression of HSL and down-regulation the expression of FAS. |
