Abstract:Objective:Recently years, the research on pathophysiologic mechanism for the brain tissue damage caused by cerebral hemorrhage is becoming a hot issue; the monoamine neurotransmitter is found that has played an improtant role in the cerebral hemorrhage secondary brain lesion. Ventricular hemorrhage which is bleeding in special parts is more likely to result in the hypothalamus、brain stem damaged. The NE contents in brain stem changed significantly, had aggravated the brain and other organs impairment.The experiment is used to observe the changes of the contents of the noradrenaline NE/NA of the brian tissue of the rat after the ventricular hemorrhage and the expressing changes of the noradrenergic neuron in the locus ceruleus, and to research the damage mechanism in the ventricular hemorrhage; thus it has pratical clinical meaning for diagnosing disease, predicting illness and instructing the treat after recovery. Method:1. preparing for IVH model:taking the right paracele as the injection point according to the《rat brain stereotaxic atlas》; making the IVH model by injecting the blood of the rat into its own brain; 2. animal grouping and processing:grouping the animals into normal group、shame operation group and the IVH group randomly; preparing the IVH model by the method of injecting their own blood in their brains for group IVH; only opening the scalp and drilling on the skull for the shame operation group; there are 118 experimental animals, specific processing:(1) neurobehavioral scoring:scoring the normal group、shame operation group and the IVH group in five time points(n=4) after operation; (2) measuring the water content in brain tissue:detecting the water content for the normal group、shame operation group and the IVH group at five time points (n=4); (3) measuring the content of noradrenalin for brain stem:taking the brain stem to measure the content of noradrenalin for the normal group、shame operation group and the IVH group after operation at five time points (n=4); (4) DHB immunohistochemistry chromoing:taking the locus ceruleus of the normal group、shame operation group and the IVH group to chromo after operation at five time points (n=4) and counted DHB positive neurons; (5) ventricular system HE chromoing:the normal group (n=4)、shame operation group(n=4), IVH group after operation at five time points (n=4),taking the ventricular system to chromo; Results:(1) neurobehavioral scoring:the scoring result of the IVH group is obviously lower than the normal group、shame operation group 6h and 1d after the operation (P<0.05); the comparison difference between three groups after injecting blood is meaningless on statistics; (2) the water content of the brain tissue in IVH group is obviously higher than the normal group and shame operation group,it begins to rise 6h after operation and gets the highest point after 1d, during the hematoma absorbing period (3d fter hemorrhage) the water content of the brain tissue is gradually lowing, at seventh day it is still higher than the contrasting group; the difference still has meaning on statistics (P<0.05); (3) the content of noradrenalin of the brain stem:the content of noradrenalin of the IVH group begins to rise gradually after 6h and get the highest point after Id, it begins to decrease 3d after injecting blood, but it is still higher than the contrasting group;the comparison differences after 6h、1d and 3d to contrasting group are with meaning on statistics (P<0.05); (4) DHB chromoing:the DBH positive neurone in the LC of the the normal group and shame operation is arranged closely, the shape between cells is unclear,the cell is in round shape, oval shape or irregular shape; the cells are multipolar neuron with cell nucleus in the middle; the chromoing color is light, the cells are extruded massively to toward the outside of the cell nucleus, there are much deeply chromoed DBH positive neurone in the locus ceruleus of the IVH group with similar form to the normal group and shame operation; the number of the positive cells and chromo are obviously increased comparing with the normal group and shame operation (P<0.05); (5) HE chromoing:the ventricular system periosteum of the normal group and shame operation is intact, there is no blood debris, and the periventricle neurone is normal, the ventricular system periosteum of the IVH group is in focal delection, there is blood debris in room; and the organization are loose and hydrops, the acidophily dead neurone can be seen. Conclusion: the rat IVH animal model made from self-blood is simple in method, easy to operate; the model is high in success rate, and it perfectly simulates the natural process of IVH of human beings and shows the similar illness change of the IVH of human beings; after the IVH, the expressing of the DBH in the locus ceruleus of the rat brain stem is improved, brain tissue nordrenalin content is increased,and has a positive correlation with the water content of the brain tissue, has a negative correlation with the neurobehavioral scoring, thus the NE is proved that it might has important effect in the brain tissue damage after the ventricular hemorrhage, and to cause other organs secondary lesion, aggravating illness.
