Vancomycin, a glycopeptide antibiotic, has been used as the first-line drug for treating serious MRSA infections, each Pharmacopoeia takes a well-defined demand to its components to regiment quality and make sure medication for patients safty and effectivity.There are some unknown components in the metabolites of vancomycin-producing strain. With the application in clinical of vancomycin, the analysis of effective components and related compounds are increasing more and more attention.This paper studies some unknown components in the the metabolites of Amycolatopsis orientalis by modern separation and purification methods, the main achievements:An unknown minor component LYV091x03 in the vancomycin products was enriched by means of vancomycin hydrolysis. The influences of different temperature,time and acid concentration on the yield of LYV091x03 in the hydrolysis were studied, also the hydrolysis conditions to LYV09lx03 were optimized and utilized:4mol·L-1hydrochloric acid,80℃and hydrolyze 2.5h could get a high relative amount of LYV091x03. According to biogenesis and chemical analysis and combined with the analytic results of other methods like macroporous adsorptive resins, medium-pressure LC method and HPLC method, the structure of LYV091x03 was identified Desamidodesvancosaminylvancomycin.The molecular weight is 1305.Although its activity in vitro is lower than Vancomycin, it is possible to used as new precursor for further synthetic use.Two unknown minor components LYV09lx01and LYV09lx02 in the vancomycin fermentation broth were studied. A specific macroporous adsorptive resinAB-8 was screened for isolation process.Through the qualitative analysis (including ultraviolet absorption spectrum,pH paper chromatography and colour reaction), The LYV091x01 and LYV091x02 were initially determined meta acids, water-solubility flavonold. Combined with the medium-pressure LC method,MS and NMR, LYV09lx01 and LYV091x02’structures were identified 4H-1-Benzopyran-4-one,5,7-[(6-deoxymannopyranosyl)oxy]-3-(4-hydroxyphenyl)and Daidzein 7-rhamnose. The physical and chemical properties of these two compounds were studied:LYV091x01 which is a new daidzin compound has a certain inhibition to Candida albicans. The changes of metabolites in different fermentation conditions of Amycolatopsis orientalis HCCB10007 were examed,includeing fermentation medium, initiate pH, temperature, dissolved oxygen.A optimized fermentation medium was obtained for LYV091x01andLYV091x02,comparied to the control medium,the yield of LYV091x01and LYV091x02 were increased by 87%and 132% respectively while other components greatly decreased, The media was optimized for future potential application.
