Purpose:Ginsenoside Re (Re) had been proved one of effective components of ginseng in medical and pharmacological studies. It was believed that ginsenoside Re content formed 23% of total ginsenosides. Therefore, it should be the representative of ginseng. However, fewer pharmacological studies on ginsenoside Re had been carried out and the effects of it on shock had not been reported. To evaluate the effect of ginsenoside Re on shock in animal models in our research, we hope to provide preclinical pharmacological experimental data.Methods:Hemorrhagic shock (HS) of rats was initiated by blood withdrawal till the mean arterial pressure (MAP) reached 40 mmHg and maintained this state for 60 min, then model animals were resuscitated by transfusion of tested substances. Systolic blood pressure (SP), Diastolic blood pressure (DP), MAP and heart rate (HR) were monitored. The blood samples were collected in 2 hour after HS respectively, to determine serum K+, Na+, Cl-, TP, ALT, AST, CK, LDH, Cr, BUN and GLU. The mortality and survival time were recorded after transfusion. And then histologis assay was performed.In mice insulin shock model, it was evaluated through recording the alive time and convulsion latency of insulin administrated animal after ginsenoside Re injection. Then serum glucose of mice in insulin shock were determined in every 10 minutes after treatment with Re.Result:Ginsenoside Re increased SP, DP and MAP significantly after transfusion. The survival time of animals in Re and (Re+PD) group were significantly longer than in model group. Six hours survival rate of Re and (Re+PD) group were significantly higher than that of model group. The survival rate of Re was higher than that of PD, and the survival of (Re+PD) group was much higher than PD group. Treatment of rats upon resuscitation with the Re and (Re+PD) reduced cardiac, liver injury and renal dysfunction. Ginsenoside Re and (Re+PD) group decreased significantly CK, LDH, K+, ALT, AST, TP, Cr, BUN, and GLU in serum.Alive time of mice in ginsenoside Re group was prolonged and serum glucose of mice in ginsenoside Re group were obviously higher than that in model group in insulin shock.Conclusion:Ginsenoside Re showed an anti-shock activity. Joint use of ginsenoside Re and PD could also significantly increase the survival rate of HS rats. It suggested that ginsenoside Re have an effect on adjuvant therapy for shock.
