Garlic is a very promising natural medicine, which has a broad charmacological effects and is more and more common in clinical applications. This review has attempted to make a bridge on high-lipid mice and the active substances of garlic—allicin and alliinase, trying to explore its lipid-lowering mechanism.When the high-lipid mice feed with 10mg/ml,5mg/ml,2mg/ml allicin and the compounding of alliinase and garlic acid(1:2), the results revealed that the serum lipoprotein levels in high-lipid mice were decreased significantly compared with the model group. It was found that TC levels had decreased by 34.5%, 28.7%, 21.4% , after treated with different concentrations of allicin , which had significantly different(P<0.01).When fed with different concentration of the compounding ,TC levels had decreased by 36.9%, 29.2%, 22.1%,which has a significant difference.The group of different dosage’s allicin raises the HDL-C content is 30.4%, 22.5% and 17.1% respectively. The mixture of alliin + alliinase raises the HDL-C content is 36.4%, 24.2% and 17.7%. The latter enhances the effect of HDL-C surpassing the former.. The group of different dosage’s allicin reduces LDL-C content is 19.0%, 12.3% and 10.8% respectively. The mixture of alliin + alliinase reduces LDL-C content respectively is 22.5%, 14.9% and 14.0% respectively. Identically, The latter reduces the effect of LDL-C surpassing the former. The allicin and the mixture of alliin + alliinase to HDL-C and LDL-C’s influence is remarkable among different groups, P<0.01, which indicates dose-dependent.The group of different dosage’s allicin enhances the SOD enzyme activity is 32.5%, 20.9% and 14.3% respectively. The mixture of alliin + alliinase enhances the SOD enzyme activity is 35.1%, 28.3% and 17.4% respectively. The trend of the garlic active material to the effect of SOD enzyme activity from mouse liver is identical with the trend of SOD enzyme activity from blood, but the SOD enzyme activity from liver is bigger slightly than the blood’s. In the liver, the ability to clear the oxygen free radical is good. The allicin and the mixture of alliin + alliinase enhance the SOD enzyme activity showing dose-dependent, and the latter to the effect of SOD enzyme activity surpasses the former.The group of different dosage’s allicin reduces the MDA content from blood is 29.9%, 27.4% and 15.5% respectively. The mixture of alliin + alliinase reduces the MDA content is 35.6%, 30.9% and 21.2% respectively. The allicin and the mixture of alliin + alliinase reduce the MDA content presenting dose-dependent. The latter is better than the forme in reducing the MDA from blood. The trend of allicin and the mixture of alliin + alliinase to reduce the MDA in liver is identical, but in the MDA content in liver is obviously lower than that of blood , which indicats that the lipin peroxidation mainly occurs in the blood .HMG-CoA reductase is the rate-limiting enzyme of cholesterol synthesis. Its activity was positively correlated with cholesterol synthesis. The different concentration of allicin (high, medium ,low) could decreased the activity of HMG-CoA reductase by 31.4 %、38.7%、44.6%. The different concention of compounding(high, medium, low) could reduced the activity of HMG-CoA reductase by 33.6 %、39.6%、46.1%. The groups between in high, medium and low doses of allicin and the compounding shows a significant different(P<0.01) by dose-independent.Product the mouse liver paraffin sections and put it in light microscopy. Most liver cells in high-lipid mice were swelling, blurring, light staining cytoplasm containing lipid droplet more or less, some cells showed vacuolar with lipid droplet fusioned, and were different to identify. The lower doses of active substance in garlic had no significant improvement. The liver cells were improved to normal ones by high dose of active substance. Compared with model group, the compounding of garlic acid and alliinase in high doses group had significant improvement in the roles of cell shape. The fat vacuoles in liver cells decreased .The nucleus were large and round and the cells were basically normal.
