| Objective:To study the effectively and safely of human umbilical cord blood stem cells transplantation by intravenous administration on hindlimb ischemia of diabetic rats.Method:1.To establish the acute hind limb ischemia on SD rats with diabetes mellitus.2. Diabetic SD rats with hind limb ischemia (n=42) were randomly allocated into six groups for transplantation. Group A (n=5):normal saline intramscular injection group, group B (n=6):normal saline intravenous administration group(by the tail vein), group C (n=8): Bone marrow stem cells intramscular injection group, group D (n=8): bone marrow stem cells intravenous administration group(by the tail vein), group E (n=8):umbilical cord blood stem cells intramscular injection group, group F (n=7):umbilical cord blood stem cells intravenous administration group(by the tail vein).3. The density of capillary was observed by Factorâ…§related antigen immunochemical stain. The expression of vascular endothelial growth factor (VEGF) in the ischemic area was detected by ELISA assays.Results:1.The secretion levels of VEGF were significantly increased after human umbilical cord blood stem cells transplantation(group E, F) than the control group (group A, B) (P<0.05),and the densities of capillary in the ischemic area were elevated (P<0.05).2. Intravenously administered stem cells (group D, F) could increase the levels of VEGF and the densities of capillary in the ischemic area than the control group (group B)(P<0.05).3. There was no significant difference between marrow stem cells transplantation group (group C, D) and umbilical cord blood stem cells transplantation group (group E, F) in ischemic area on capillary densities and VEGF levels (P> 0.05).4. No significant difference was observed between stem cells intramscular injection group(group C, E) and intravenous administrator group (D, F group) on capillary density and VEGF levels (P> 0.05).Conclusion:â‘ The secretion level of VEGF and density of capillary in ischemic area were significantly increased after human umbilical cord blood stem cells transplantation,â‘¡Intravenously administered human umbilical cord blood stem cells could increase the level of VEGF and the density of capillary in the ischemic area,â‘¢Intravenously administered human umbilical cord blood stem cells may be a new choice for the treatment of diabetic food. |
