Objective:To investigate the Water Extracts of Zuojin Prescription (WEZP) induced apoptosis in SGC-7901 cells, and analyse the potential material basic of the effect of WEZP To explore the rational way of using WEZP effectively and correctly in Patients with gastric cancer.Method:The time and dose influence of WEZP was tested by MTT assay, PI staining flow cytometry, the apoptotic rate was investigated by FCM and the morphology was observed by Hoechst 33258 staining. Then Anti-cancer drug filtration from Berberine, Palmatine,Jatrorrhizine, Evodiamine, Rutecarpine etc, and those five kinds of alkaloids in Zuojin Prescription. Finally, western blotting was performed to explore the bcl-2, bax gene and protein expression in SGC-7901.Results:1.WEZP does not stimulates cell growth obviously in vitro. The other way round, it may act as a SGC-7901 cell grow inhibitor. The inhibited cell proliferation and induced cell apopotosis in a dose-dependent and time-dependent manner.2. Five different alkaloids in WEZP, the tumor inhibited affection descending from Berberine, Palmatine, Evodiamine, Rutecarpine, to Jatrorrhizine.3. Not only Berberine but Palmatine can reduce the bcl-2 gene expression and increased gene expression of bax.Conclusions:WEZP could significantly inhibit the growth of SGC-7901 cells and induce apoptosis reduce the bcl-2 gene expression and increased gene expression of bax may be one of apoptosis mechanism.
