Piperazine is commonly used as a kind of basic groups, basic interval units or limitative conformation of ethylenediamine units in the pharmaceutical chemistry research. It is a dual-function compound with two secondary amines, which has fewer side effects than the primary amine during the chemical reactions. It is easy to dissolve in the water and its alkalinity is moderate. When it is introduced into the drug, it can effectively regulate the fat and water distribution coefficient and pKa value, forming muti-hydrogen bonds or ionic bonds. Monosubstituted Piperazine, because of its unique structure, has become the key intermediate in the numerous medicine synthesis. Therefore, the synthesis of monosubstituted piperazines has the very important pharmacy value and the economic value.The sulfonamide is also called the sulfanilamide, which is a quite commonly used kind of medicine. Its main role is to inhibit multiplication of bacteria to achieve the purpose of antibacteria, not to kill the bacteria directly. The antibacterial spectrum of this kind of medicine is broad, and it has the inhibitory action to many kinds of coccuses and certain bacilli.Although the sulfanilamide medicine has many merits, its fatal shortcoming has restricted its application seriously. Meanwhile, along with the development of antibiotic and other chemotherapy medicine, the clinical application of sulfonamides has the reduced tendency. The piperazine ring has the excellent physical and chemical properties, and is also the medicine in itself, After introduced into the sulfa drugs, it can not only increase the drug efficacy group, but also improve their physical and chemical properties.On the basis of extensively reading literatures about the research and application of sulfa piperazines at home and abroad, this thesis designed and synthesized a series of new types of sulfanilamide analogs containing piperazine ring, and discussed their anti-microbial activities initially. This research including six major parts:(1) The preparation of 4-(4- substituentpiperazin-1-ylsulfonyl)benzenamine;(2) The preparation of 4-(piperazin-1-ylsulfonyl)benzenamine;(3) The preparation of 4-(4-(4-(4-aminophenylsulfonyl)phenyl)piperazin-1-ylsulfonyl)benzena-mine;(4) The preparation of p-aminobenzolic acid derivatives;(5) The preparation of amino acid derivatives;(6) The determination of biological activity of above compounds.
