Objectives To observe the effect of Yang xue qing nao Ke Li on the expression of brain-derived neurotrophic factor BDNF and other related proteins in CA1 area of hippocamps of vascular dementia rats.To explore the possible mechanism of Yang xue qing nao Ke Li in the treatment of VD.Methods 1.SD rats were randomly divided into sham operation group, vascular dementia model group and Yang xue qing nao Ke Li treatment group. After the success of the model, the principle of randomization was divided into 1 W, 2 W, 4 W and 8 w 4subgroups, each subgroup had 6 rats.among which, 3 were used for Transmission Electron Microscope, 6 rsts were used for immunohistochemistry, and the let 6 rats were detected by Western blotting. 2.The Vascular Dementia rat model was prepared by modified Pulsineli’s four-vessel occlusion. 3.Rats in the three groups were administered orally, 1 times a day. sham operation group: After the bilateral common carotid artery of the rats were separated, the ischemic treatment was not done. Only normal saline was Gavaged(10ml/kg.d). model group: After the successful preparation of vascular dementia model, Gavage normal saline 1w, 2 w, 4 w, 8 w(10ml/kg.d). treatment group: Referring to the method of reported by Tao Tao et al, The Yang xue qing nao ke li is configured with distilled water suspension(concentration of 0.32g/ml, 10ml/kg.d), Gavage Yang xue qing nao ke li(10ml/kg.d) 1w, 2 w, 4 w, 8 w after model successfully administered.4.The changes of ultra micro-structure of neural cells were observed by transmission Electron Microscope. The expression of BDNF, P38 MAPK, GSK-3β and β-cateninin CA1 Area of hippocampus of Vascular Dementia were detected with immunohistochemi--cal analysis and western blotting method. 5.e observe tissue slices with Microscope camera OLYMPUS(400′),then analysis them by Med Motic 6.0 digital medical image analysis system.The results of western blotting were analyzed with Image J.6. 6.Measure--ment date are presented as mean±SD( sx ±), application SPSS 17.0 statistical software for date analysis at last, using anova design analysis of variance within the group, P<0.05 for the difference has statistical significance.Results 1.In terms of cognitive function, Compared with the sham operation group, the number of crossing platform was significantly reduced, and the difference was statistical-ly significant(mean P<0.01), Compared with the model group, the number of cross platform was increased, and the difference was statistically significant. 2.In the sham group, The structure of CA1 neurons in the rat hippocampus is complete and the boundary is clear. Nuclear morphological rules, nuclear membrane was complete, there are rich organelles in cytoplasm, the structure is normal;In the model group, most of the neurons in CA1 region pyknosis, nuclear dissolution, heterochromatin margination,mitochondria swelling. In the treatment group, the distribution of chromatin was wellproportioned, and normal mitochondrion and other organelle were observed. 3.In the sham group, the CA1 P38 MAPK, GSK-3β, BDNF and β-catenin protein expression were found in the hippocampus of rats at different time points. In the model group, at each time point of P38 MAPK, GSK-3βand BDNF expression increased, 1 weeks when there is a multi expression, 2 weeks that have large quantities of expression, peaked at 4 weeks,began to decline but still higher at 8 weeks. Compared with sham group, the expression of P38 MAPK, GSK-3βand BDNF in CA1 area of hippocampus of vascular dementia rats were significantly increased in model group at different time points(all P<0.01). The expression of β-catenin in CA1 area of hippocampus of vascular dementia rats were increased in model group at different time points, compared with sham group, the expression of β-catenin were no significantly(P >0.05). The protein expression of BDNF,GSK-3βand β-catenin in treatment group were significantly increased at different time points. Compared with model group, the expression of P38 MAPK in CA1 area of hippocampus of vascular dementia rats were significantly decreased(all P<0.01).Conclusions Yang xue qing nao KeLi can improve the cognitive function of vascular dementia,improve the ultrastructure of neurons in hippocampus CA1 area of vascular dementia rats;The mechanism may be related to up regulation of the expression of BDNF,GSK-3β and β-catenin,Inhibiting the expression of P38 MAPK protein.
