Objective : To investigate the pathologic characteristics of CART1( cartilage hemeprotein 1) in breast cancer and adjacent tissues. To investigate the effect of exogenous small interference RNA-mediated silencing of the CART1 gene on cell biology behavior in human breast cancer cell MDA-MB-231.Method:The relationship between CART1 and breast cancer patient’s age 、tumor size、pathological type、pattern of organization and clinical stag were detected by immunohistochemistry. We designed and chemically synthesized siRNA targeting CART1 or control, and then siRNA was transfected into MDA-MB-231 cells using lipofectamine. The suppression effect of siRNA on CART1 expression was determined by quantative Real-time PCR and Western Blot, respectively. The effects of CART1 knockdown on MDA-MB-231 cells invasion, proliferation and cell cycle were detected by Matrigel invasion assay, cell counting kit and flow cytometry.Result : In 62 cases of breast cancer,CART1 was highly expressed(87.1%) in 54 cases, and CART1 was not expressed or lowexpressed in the adjacent tissues of 8 cases.CART1-siRNA could significantly suppress the expression of CART1 mRNA and protein as compared with the control group. MDA-MB-231 cells transfected with CART1 siRNA had lower levels of invasion proliferation capacity compared with the control group.Conclusion:High expression of CART1 in breast cancer.Depletion of the CART1 gene could significantly inhibit cell invasion and proliferation and induce cell cycle arrest in S phase in human breast cancer cell line MDA-MB-231.
