| ObjectiveTo measure the effect of using 3-D polyporous small intestinal submucosa for bladder regeneration in a rabbit model.MethodsWith physical and chemical method, 3-D polyporous small intestinal submucosa(SIS) was made; A total of 24 male New Zealand white rabbits were randomly divided into three groups respectively treating partial cystoplasty, and treated with three types of SIS for bladder reconstruction,including Control group: without peroxyacetic acid(PAA) treated SIS(n=8),PAA treated group:PAA treated SIS(n=8), and PAA and serum treated group: PAA treated SIS and with 100% fetal bovine serum infiltration(n=8). Three groups in the preoperative and postoperative 4weeks to measure bladder capacity, and in postoperative 4 weeks, gross observation and histological examination were performed to evaluation of bladder regeneration.Results4 weeks after partial cystoplasty,tissue adhesion of Control groupã€PAA treated group and PAA and serum treated group with the surrounding is reduced in turn, and the rate of stone formation is 2/6 〠2/7 〠1/7respectively.Compared with preoperative itself 4 weeks later, bladder capacity of three group were all narrowed and the capacity increased in turn in three groups,comparison between groups(P<0.05).Histological evaluation revealed that Urothelial cells regeneration of three groups is good;compared with Control group,PAA group and PAA and serum treated group is better, With no significantly difference between the marginal zone and the central zone. However,smooth muscle regeneration are not ideal,the marginal zone is better than the central one,comparison between groups( P<0.05); The regeneration area of blood vessels increases in turn in three groups, the comparison between the three groups was statistically significant(P < 0.05).Conclusion3-D polyporous small intestinal submucosa treated with PAA and fetal bovine serum has the ability of promoting the bladder tissue regeneration,regarding as an ideal alternative to repair materials. |
