Objective: To explore the efficacy and safety of 125 I seed implantation for treatment of neuroblastoma animal models and provide research basis for later clinical application of this technology.Methods: Cultivated neuroblastoma SK-N-SH cell line, to establish45 of nude mouse models of neuroblastoma, divided125 I group, control group and blank group, 15 in each group. According to the formula implanted the corresponding number of 125 I radioactive seeds in 125 I group’s nude mice, every 3 days measured the tumor diameter a, short diameter b.Every 9 days handled of each groups of 5 mice, weighed the weight heart,liver, spleen, kidney, computed organ index and the tumor inhibition rate of weight and volume. Detected expressions of apoptosis and proliferation of tumor tissue and peritumoral tissue(around the tumor about 1-2 cm) by Western blot and immunohistochemical staining. For the detection of endocrine function by ELISA kit. The apoptosis of tumor tissue was detected by TUNEL.Results: Efficacy index:(1) 125 I group tumor volume inhibition rate at9 th day reached the maximum 73.55% and trended downward(P < 0.05).Tumor weight inhibition rate at 9th, 18 th, 27 th respectively were: 38.57%,29.89%, 26.48%, and trended downward at 9th to 18 th day(P < 0.05), had no difference at 18 thto 27thdays(P > 0.05). Tumor weight and volume inhibition rate had no difference in control group and blank group.(2) In tumor tissue,the expression of caspase-3 in125 I group were the highest thanthat in control group and blank group(P < 0.05). the expression of PCNA in 125 I group were the lowest than that in control group and blank group by Western blot and immunohistochemistry(P < 0.05).(3) The apoptosis of tumor tissue in 125 I group were the highest than control group and blank group by TUNEL(P < 0.05).Safety index:(1) the organ indexes, blood indicators and endocrine indexes in all groups had no significant difference(P>0.05).(2) Had no significant bone marrow suppression phenomenon in 125 I group and no significant difference with control group and blank group.(3) The expression of PCNA and caspase-3 in 125 I group were no significant difference with control group and blank group by Western blot and immunohistochemistry in peritumoral tissue.Conclusion:(1) 125 I seeds has obvious effect in neuroblastoma nude mice..(2)125 I seeds implantation is one of the ways in which close radiotherapy,the experiment proves the therapeutic dose was safety in neuroblastoma nude mice.
