Objective: ITRAQ-based tandem mass spectrometry was employed to quantify proteins secreted by AIS patients and the controls,and some of differentially expressed proteins were validated by ELISA.Methods: The serum samples from AIS patients and the controls were concentrated. With iTRAQ and LC-MS/MS analysis,we screen out differentially expressed proteins, furthermore, AHSG and HPX were selected for validation by ELISA.Results: We found 29 proteins(9 up-regulated proteins and 20 down-regulated proteins)significantly alter in AIS patients. DAVID analysis and STING network suggest that HPX, AHSG are acute inflammatory reaction related protein. ELISA showed AHSG(AIS:424.0±113.3 mg/L,the controls:526.9±124.0,P=0.001)and HPX(AIS:810.9±164.1 mg/L,the controls : 954.8±85.6 mg/L, p<0.001)serum levels significantly decreased in AIS subjects compared to the controls, which were consistent with the results of mass spectrometry.Conclusions: The concentration of AHSG and HPX in AIS patients serum showed an decreasing trend.The deregulated proteins may be useful as potential therapeutic targets or biomarkers for acute ischemic stroke.
