The Research Of Expression Of MMP-2, 3, 9, 11, 14 And Function MMP-11 In Breast Carcinoma

Objective: To analyse the expression of MMP-2,3,9,11 and 14 and their possible clinical significance in breast carcinoma patients and detect To investigate the effect and mechanism of MMP-11 on breast carcinoma cell migration and invasion.Methods: MMP-2,3,9,11 and 14 were detected by immunohistochemical technique in 161 invasive breast carcinoma tissues,39 adjacent tissues of breast cancer.The relationship between the expression of MMP-3,9,11 and 14 in invasive breast carcinoma tissues and the clinicopathological characteristics of breast patients,menopausal status,tumor size,histological grade,lymph node metastasis status and TNM stage were analyzed. SiRNA was used to knockout MMP-11 in breast carcinoma cell line MDA-MB-231 and Transwells were used to evaluate changes in migration ability and invasion ability.Western-blot was used to detect MMP-11 before and after knockout.Results: MMP-3, 9, 11 and 14 in invasive breast carcinoma tissues were high expressed,but in 39 adjacent tissues of breast cancer,These four proteins were feeble even unexpressed. MMP-2 in invasive breast carcinoma tissues and 39 adjacent tissues of breast cancer were high expressed. There were some differences between the expression of these four MMPs and the clinicopathological characteristics in invasive breast carcinoma, and having a certain clinical significance.After knockout of MMP-11,the expression of both proteins decreased in MDA-MB-231 cell line and experiments show that the cell’s migration and invasion abilities are significantly weakened.Conclusion: The differential expression and different correlation of MMP-2, 3, 9, 11 and 14 in breast carcinoma Which were closely lined with the occur and development of breast carcinoma.and knockout of MMP-11 The inhibition of breast carcinoma cell’s migration and invasion.They could be potential prognostic markers and treatment targets for of breast carcinoma.
