The AMP-activated protein kinase(AMPK) is an important cellular energy monitor. AMPK is activated under low energy state and stress. It plays a huge role in cellular energy homeostasis. Recent studies found that AMPK also has important role in the process of pathological physiology of development of cancer, inflammatory reaction and apoptosis. Our research team previously discovered that cellular energy state was closely correlated with the progression of lipopolysaccharide/D-galactosamine(LPS/D-Gal)-induced acute liver injury. In order to explore the role of AMPK in LPS/D-Gal-induced liver injury. We used the AMPK activator A-769662 and the AMPK inhibitor CC in this model and explored the mechanism from the aspect of liver injury and inflammation and apoptosis. Our evidence indicated that inhibitory of AMPK reduce the liver injury and apoptosis, activated of AMPK anabatic the liver injury and apoptosis and then we tested the activation of P53 and JNK whose activity along with the activity of AMPK but P53.The result show that AMPK activator A-769662 can aggravate LPS/D-Gal-induced acute liver injury and the mechanism might involve with JNK signal path. These information revealed that AMPK maybe exert detrimental roles in LPS/D-Gal induced liver damage and inhibit the activity of AMPK may become a new orientation for therapy of inflammation and apoptosis-related liver disease.
