Objective: To detect the microdeletions of Y chromosome Azoospermia factor(AZF)region in infertile males.We aim to discuss the relationship between Y chromosome microdeletion and male infertile, and the relevance of the microdeletions in AZF region and sex hormone levels.Method: 127 cases of non-obstructive azoospermia and 23 cases of severe oligozoospermia were enrolled. Multiplex polymerase chain reaction(PCR) was used to detect the microdeletions in AZF region,the levels of gonadal hormone were detected by Electrochemiluminescence immunoassay(ECLIA).Results:1、Among the 150 male infertile patients, 19(12.7%) were found have microdeletion in AZF regions.In 2 of the 23 severe oligozoospermia patients have microdeletion and the incidence of non-obstructive azoospermia patients was 13.4%.Of 19 patients with microdeletions in AZF region,2 patients had only one deletion in AZFb regions(10.5%),1 patient had deletions in AZFb combined with AZFc(5.3%), 4patients had deletions in three AZF regions: AZFc combined with AZFb and AZFd(21.0%), 12 patients had deletions in AZFb combined with AZFc(5.3%).In 15 STSs, apart from s Y82、s Y84、s Y86 the other twelve STSs were all found deletions of AZF region.2、The levels of T in non-obstructive azoospermia group and severe oligozoospermia group were same as the control group, there were no statistical significance( P >0.05);but there were statistical significance in the levels of FSH and LH between non-obstructive azoospermia group and control group(P<0.05). the levels of FSH in the patients with AZFb+c+d deletions were much greater than patients with AZFb or AZFb+c or AZFc+d deletions and non-AZF delections patients, there were statistical significance among them.Conclusion:1 、 The result showed that the incidence of microdeletions in AZF region is12.7 %,in 15 of the 127 non-obstructive azoospermia patients have united microdeletion.It is suggested that the united microdeletions in AZF regions may be an important cause of non-obstructive azoospermia.2、There was a certain relationship between serum FSH levels and AZF region gene microdeletion, AZF region gene microdeletion can causes increased in serum FSH level.3、There is a clinical significance in detecting the AZF region gene microdeletion between non-obstructive azoospermia and severe oligozoospermia patients.
